Abstract

Disentri basiler adalah suatu infeksi akut pada usus besar yang disebabkan oleh Shigella dysenteriae. Kasus multi resistensi Shigella dysenteriae terhadap antibiotik ditemukan di seluruh dunia dan timbul sebagai akibat penggunaan antibiotik yang tidak rasional salah satunya di Indosesia. Potensi penggunaan tanaman sebagai pengobatan sudah sejak zaman dahulu digunakan. Berdasarkan etnomedicine daun kastuba telah digunakan sebagai tanaman obat untuk mengatasi disentri. Oleh karena itu, diperlukan alternatif pengobatan dalam mengatasi bakteri Shigella dysenteriae salah satunya dengan meggunakan tanaman obat. Tujuan dari penelitian ini untuk mengetahui efek antibakteri ekstrak daun kastuba terhadap bakteri Shigella dysenteriae secara in vitro. Metode yang digunakan adalah metode dilusi agar untuk menentukan Kadar Hambat Minimum. Ekstrak daun kastuba dibuat dengan cara maserasi menggunakan etanol 70%. Konsentrasi ekstrak daun kastuba yang digunakan adalah 0%; 0,2%; 0,3%; 0,4%, dan 0,5 % v/v, sedangkan konsentrasi Shigella dysenteriae adalah 104 CFU/ml. Hasil penelitian ini menunjukkan bahwa ekstrak dari daun kastuba mempunyai kandungan flavonoid, saponin, dan tannin, memiliki aktivitas antibakteri terhadap pertumbuhan bakteri Shigella dysenteriae. Kadar Hambat Minimum diketahui terdapat pada konsetrasi 0,5 % v/v. Analisis data menunjukan perbedaan bermakna antara konsentrasi ekstrak dengan jumlah koloni yang tumbuh pada kelompok sampel (Kruskall Wallis, p < 0,05). Uji korelasi spearman menunjukan adanya hubungan yang erat antara konsentrasi ekstrak dengan jumlah koloni yang tumbuh (Korelasi, R = -0,909; p = < 0,05). Kesimpulan dari penelitian adalah ekstrak daun kastuba mampu menghambat pertumbuhan Shigella dysenteriae dengan KHM 0,5% v/v.

English Abstract

Bacillary dysenteriae is an acute infection in colon which is caused by Shigella dysenteriae multiresistant cases of Shigella dysenteriae toward antibiotics was found in the worldwide and it was appear because of irrational usage of antibiotics which also happen in Indonesia. Potential use of plants as medicine has been known since long times ago. Based on etnomedicine kastuba leaf has been used as a medicinal plant to treat dysentery. Therefore, kastuba leaf proposed to have a potensial as an alternative treatment to treat the infection of bacteria Shigella dysenteriae. The purpose of this research was to find antibacterial effect of kastuba leaf extract for in vitro Shigella dysenteriae. The tube dilution method was used for knowing Minimum Inhibitory Concentration. kastuba leaf extract was made by maceration with 70% ethanol. The extract concentrations used were 0%; 0.2%; 0.3%; 0.4%; and 0,5% v/v, in the other hand concentration of Shigella dysenteriae was 1x104 CFU/ml. The result showed that extract of kastuba leaf contain flavonoids, saponins, and tannins which had antibacterial activity against the growth of Shigella dysenteriae. Minimal Inhibition Consentration was in 0,5 % v/v of concentration. Data analysis showed that the significant differences between extract concentration with colony which grow in sample group (Kruskall Wallis, p < 0.05). Spearman correlation test show there is a strong relationship between extract concentration with the amount of growing colony (Correlation, R = -0,909; p = <0.05). As a conclusion, the research shows that kastuba leaf extract can impede the growth of Shigella dysenteriae with MIC on 0,5% v/v.
