Abstract

Shigella dysenteriae merupakan spesies yang paling sering menyebabkan kejadian endemik yang parah. Hal ini disebabkan karena hanya dengan 10 basil Shigella dysenteriae dapat menimbulkan penyakit pada beberapa individu, sedangkan serotipe lain membutuhkan 100-200 basil. Shigella dysenteriae juga cepat menjadi resisten terhadap banyak obat antimikroba sehingga menimbulkan masalah terapi yang sulit. Daun pepaya (Carica papaya L) mempunyai beberapa senyawa aktif yang larut dalam air. Tujuan penelitian ini adalah untuk mengetahui hubungan antara konsentrasi ekstrak etanol daun papaya (Carica papaya L) terhadap pertumbuhan koloni bakteri Shigella dysenteriae secara in-vitro dan untuk mengetahui Kadar Hambat Minimum (KHM) dan Kadar Bunuh minimum (KBM) ekstrak etanol daun papaya terhadap Shigella dysenteriae. Sampel daun diekstraksi menggunakan pelarut etanol 96% dengan metode perkolasi yang dilanjutkan dengan proses evaporasi, digunakan lima macam konsentrasi ekstrak daun pepaya (Carica papaya L), masing-masing 12%, 14%, 16%, 18% dan 20% v/v serta konsentrasi 0% sebagai pembanding atau kontrol Bakteri (KK). Dari hasil uji dilusi tabung dapat dilakukan pengamatan terhadap tingkat kekeruhan larutan ekstrak daun pepaya untuk menentukan Kadar Hambat Minimal (KHM). Analisa data yang digunakan adalah one way ANOVA. Analisis data menggunakan uji one way ANOVA dan korelasi regresi pada taraf kepercayaan 95% (α = 0,05). Hasil penelitian menunjukkan bahwa terdapat perbedaan jumlah koloni pada berbagai perlakuan konsentrasi ekstrak daun pepaya (Carica papaya L). Kesimpulan pada penelitian ini adalah ekstrak etanol daun papaya memiliki efek antimikroba terhadap bakteri Shigella dysenteriae secara in vitro dengan KHM 18% dan KBM 20%.

English Abstract

Shigella dysenteriae is the most frequent cause of severe endemic occurrence. This is because with just 10 bacils of Shigella dysenteriae may cause disease in some individuals, whereas other serotypes requires 100-200 bacils. Shigella dysenteriae is also fast becoming resistant to many antimicrobial drugs are giving rise to problems of difficult therapy. Papaya (Carica papaya L) has a number of active compounds that are soluble in water. The purpose of research is to know the relationship between concentration of ethanol extracts of papaya leaf (Carica papaya L) against the growth of Shigella dysenteriae in-vitro and to know Minimum Inhibitory Concentration (MIC) and Minimum Bactericide Concentration (MBC) ethanol extracts of papaya leaf against Shigella dysenteriae. Samples were extracted using a solvent of ethanol 96% by percolation method which continued with process of evaporation, used five different concentrations of extracts of papaya leaf (Carica papaya L), respectively 12%, 14%, 16%, 18% and 20% v/v and 0% concentration as a comparison or control the bacteria (KK). From the results of dilution tubes test can be made observation of turbidity levels of aqueous extract of papaya leaves to determine Minimal Inhibitory Levels (MIL). Analysis of the data used is one way ANOVA. Data analysis using a one way ANOVA test and correlation regression on a 95% confidence level (α = 0,05). The results showed that there were differences in the number of colonies at various treatment concentrations of extract of papaya leaf (Carica papaya L). Conclusions on the research is the papaya has antimicrobial effects against Shigella dysenteriae in vitro with MIC 18% v/v and 20% v/v of MBC.
