Abstract
Penurunan kadar estrogen pada wanita menopouse mengakibatkan peningkatan inflamasi pada jaringan periodontal sehingga memiliki dampak terjadinya peningkatan resorpsi tulang jaringan periodontal. Selain itu, penurunan kadar estrogen tersebut juga memicu penurunan sintesis fibroblas pembentuk kolagen sehingga dapat menyebabkan kehilangan perlekatan jaringan periodontal. Kedua dampak tersebut mengakibatkan terjadinya pelebaran ruang ligamen periodontal. Menurut penelitian sebelumnya, daun ciplukan (Physalis minima L) dengan unsur physalin yang dikandungnya diduga merupakan tumbuhan yang bersifat fitoestrogen karena memiliki efek estrogenik. Penelitian ini bertujuan untuk mempelajari efek pemberian ekstrak metanol daun ciplukan terhadap perbaikan ruang ligamen periodontal tikus wistar putih pasca ovariektomi. 24 ekor tikus dibagi secara acak menjadi 6 kelompok yaitu tikus normal (N), tikus ovariektomi 4 minggu (K1), tikus ovariektomi 8 minggu (K2), tikus ovariektomi 8 minggu yang diberi ekstrak metanol daun ciplukan pada akhir 4 minggu pertama dengan dosis berbeda (P1=500, P2=1.500, P3= 2.500 mg/kgBB). Sediaan histopatologis digunakan untuk menghitung lebar ruang ligamen periodontal. Hasil uji statistik menunjukkan terdapat perbedaan lebar ruang ligamen periodontal antara enam kelompok yang berbeda (ANOVA=0,000, P<0,05) dan terdapat korelasi yang kuat dengan arah negatif antara dosis ekstrak metanol daun ciplukan dengan lebar ruang ligamen periodontal tikus wistar pasca ovariektomi (r = -0,848, p<0,05). Kesimpulan dari penelitian ini adalah ekstrak metanol daun ciplukan (Physalis minima L.) dapat menurunkan lebar ruang ligamen periodontal tikus wistar putih pasca ovariektomi yang menandakan terjadinya perbaikan ruang ligamen periodontal pada tikus wistar pasca ovariektomi.
English Abstract
Decreasing level of estrogens on menopausal women may cause increasing of periodontal tissue inflammation which an impact on increasing bone periodontal tissue resorption. Besides, it also triggered the decreasing of fibroblast synthesis formed collagen which can cause periodontal tissue attachment loss. Such impacts cause widening of periodontal ligament space. Based on the previous studies, ciplukan leaf (Physalis minima L.) with its physalin element is indicated as a plant which has phytoestrogen characteristic since it has estrogenic effects. The purpose of this present study is to determine the effect of ciplukan leaf methanol extract administration on the repair of periodontal ligament space on rats after ovariectomy. 24 rats were randomly assigned into 6 groups: normal rats (N), 4 week ovariectomy treated rats (K1), 8 week ovariectomy treated rats (K2), and 8 week ovariectomy treated rats that were administered with the ciplukan leaf methanol extract at the end of the first 4 weeks during ovariectomy administration with three different doses (P1= 500, P2=1.500, P3=2.500 mg/kgBB). The results of statistical tests indicate that there are differences in the periodontal ligament space width between six different groups (ANOVA=0,000, p<0,05) and there is significant correlation with negative line between the dose of ciplukan leaf methanol extract and the periodontal ligament space width(r = -0,848). The conclusion shows the ciplukan leaf methanol extract (Physalis minima L.) proved it can narrow the periodontal ligament space width of wistar rats after ovariectomy which indicated as reparing periodontal ligament space on rats.
