Abstract
Avulsiadalahbentukcederatraumatikgigikelaslima yang didefinisikansebagaikeluarnyaseluruhgigidarisoketakibat trauma.Penangananuntukkasusavulsiadalahdengancaramelakukanreplantasisecepatmungkin,jikatindakanreplantasitidakdapatdilakukansecepatnyasebaiknyagigi yang avulsidisimpandalamsuatu media penyimpanan. Media iniakanmenjagasupayaselfibroblas di sekitarligamen periodontal tidakcepatmati.Penelitianinibertujuanuntukmengetahuiefektivitas air kelapasebagai media simpandalammenjagaviabilitasselfibroblas (in vitro). Jenis penelitian ini adalah eksperimental in vitro dan rancanganRandomizedPost Test Only Control Group Design. Dimana obyek perlakuan dibagi menjadi 6 kelompok yaitu, I,II,III,IV (sebagai perlakuan) dan V, VI (sebagai kontrol media dankontrolsel) secara random dan diambil datanya setelah dilakukan perlakuan. Sampel yang digunakan adalah kultur sel fibroblas. Sampel diinkubasi selama 24 jam kemudian media kultur diganti dengan media simpan yaitu air kelapa. Jumlah sel yang hidup dalam microplate di hitung dengan ELISAreader menggunakan MTT Assay. Hasil dari penelitian didapatkan air kelapaefektifdalammempertahankanselfibroblashingga 6 jam (103.0355594%) danefektifbiladigunakansebagai media simpan.
English Abstract
Avulsion is fifth type of dental traumatic injury that is defined by coming out tooth out of its socket becaused of trauma. Immediately replantation is the best treatment of avulsed tooth but if it cant be done, the tooth should be stored in a storage media to keep periodontal ligament cell alive. The purpose of this research is to determine effectivity of coconutwateras storagemedia for avulsedtoothviability of fibroblast culture cell (in vitro).This study is experimental research (in vitro) with Randomized Post Test Only Control Group Design. Experimental object group divided into four group I,II,III,IV which data is randomly taken after getting treatment. Sample using fibroblast culture cell. First of all, sample is incubated for 24 hours long then the culture media is displaced by storage media which will be tested, that is coconut water. Living Fibroblast culture cell in microplate is counted by ELISA reader using MTT Assay. The result shows that coconut water is an effective in preservating fibroblast cell up to 6 hours (103.0355594%)and effective if used as storage media for avulsed tooth.
