Abstract

Shigella dysenteriae merupakan spesies Shigella yang dapat menyebabkan penyakit Shigellosis berat dan menyebar cepat sehingga terjadi epidemi. Shigella dysenteriae cepat menjadi resisten terhadap banyak obat antimikroba sehingga menimbulkan masalah terapi yang sulit. Seledri (Apium graveolens L.) mengandung beberapa senyawa aktif yang dapat larut dalam air, diantaranya flavonoid (apigenin, isoquercetrin), saponin, dan tanin yang diduga memiliki aktivitas antimikroba. Penelitian ini bertujuan untuk mengetahui pengaruh dekok seledri (Apium graveolens L.) terhadap pertumbuhan Shigella dysenteriae secara in vitro. Sampel diperoleh dari isolat di Laboratorium Mikrobiologi FKUB. Konsentrasi akhir dekok yang dipakai yaitu 25%, 27,5%, 30%, 32,5%, dan 35% . Metode yang digunakan adalah metode dilusi tabung. Pada penelitian ini Kadar Hambat Minimal (KHM) terdapat pada konsentrasi 30% dan Kadar Bunuh Minimal pada konsentrasi 35%. Hasil statistik Kruskall-Wallis menunjukkan terdapat perbedaan yang signifikan pada perubahan konsentrasi dekok seledri terhadap jumlah koloni Shigella dysenteriae (p<0,05). Uji korelasi menunjukkan adanya hubungan yang erat antara konsentrasi dekok dengan jumlah koloni (Spearman, R = -0,988: p < 0,05). Berdasarkan penelitian ini, dapat disimpulkan bahwa dekok seledri mempunyai efek antimikroba terhadap Shigella dysenteriae.

English Abstract

Shigella dysenteriae is one of the Shigella spesies that can cause serious Shigellosis disease and spread fast so that can be epidemic. Shigella dysenteriae is quickly becoming resistant to many antimicrobial drugs and lead to a difficult therapeutic problems. Celery (Apium graveolens L.) have some active water soluble compund, such as flavonoid (apigenin, isoquercetrin), saponin, and tanin that suspected have an antimicrobial effect. This research’s goal is to identify the effect of celery’s decocta against the growth of Shigella dysenteriae in vitro. The sample is obtained from human isolate at Microbiology Laboratory of Faculty of Medicine Brawijaya University. The final concentration of decocta used was 25%, 27,5%, 30%, 32,5%, dan 35% . The method used was tube dilution test. In this research, Minimum Inhibitory Concentration (MIC) contained in concentration of 30% and the Minimum Bactericide Concentration (MBC) is in concentration of 35%. The result of Kruskall-Wallis statistics showed there is a significant difference in the change of celery’s decocta concentration againts the amount of Shigella dysenteriae colony (p < 0.05). The correlation test shows there is a strong relation between the decocta concentration and the colony amount (Spearman test, R = -0,988: p < 0,05). It can be concluded that the celery’s decocta has an antimicrobial effect against Shigella dysenteriae.
