Abstract
Escherichia coli adalah bakteri yang umumnya dapat ditemukan di usus manusia dan hewan berdarah panas lainnya. Sebagian E. coli tidak berbahaya, tetapi sebagian lainnya dapat menyebabkan penyakit-penyakit yang berat bahkan mematikan, seperti diare, infeksi saluran kemih, sepsis, dan meningitis. Penggunaan tanaman untuk pengobatan telah dikenal masyarakat sejak lama, tetapi masih diperlukan penelitian ilmiah untuk membuktikan dan mempertanggungjawabkan penggunaan tanaman obat tersebut. Penelitian ini bertujuan untuk mengetahui potensi antibakteri ekstrak daun beringin (Ficus benjamina L.) terhadap E. coli secara in vitro. Metode yang digunakan dalam penelitian ini adalah dilusi tabung. Isolat E. coli diperoleh dari Laboratorium Mikrobiologi FKUB. Daun beringin yang telah diekstraksi menggunakan cara maserasi dengan pelarut etanol 96% dibuat dalam beberapa konsentrasi: 13%, 13,5%, 14%, 14,5%, dan 15%, kemudian ditambahkan 1 ml suspensi bakteri E. coli ke dalam masing-masing tabung. Tabung diinkubasi selama 18-24 jam pada suhu 37oC kemudian diamati kekeruhannya. Hasil dilusi tabung ditanam pada medium NAP, diinkubasi, dan diamati pertumbuhan jumlah koloninya. Penelitian diulang empat kali. Dari hasil pengamatan, dapat ditentukan Kadar Hambat Minimal (KHM) sebesar 13,5% dan Kadar Bunuh Minimal (KBM) sebesar 15%. Uji statistik Kruskal-Wallis menunjukkan nilai signifikansi p<0,05. Oleh karena itu, dapat disimpulkan bahwa ekstrak daun beringin (Ficus benjamina L.) memiliki efek antibakteri terhadap E. coli secara in vitro.
English Abstract
Escherichia coli is a common bacteria found in humans’ and other hot blooded animals’ large intestines. Some of them are not dangerous, but some others can cause serious diseases, such as diarrhea, urinary tract infection, sepsis, and meningitis. People are already familiar with the using of plants to cure for a long time, but scientific researches are still needed to prove and to take responsibility of it. This research was conducted to find out the antibacterial potency of weeping fig (Ficus benjamina L.) leaves toward E.coli in vitro. The method used in this laboratory experimental research was tube dilution. The sample of E. coli was taken from Microbiology Laboratory of Medical Faculty of Brawijaya University. The leaves were extracted using maceration method with 96% ethanol as the solvent, then divided into some concentrations: 13%, 13,5%, 14%, 14,5%, and 15%, then 1 ml suspension of E. coli was added into each tube. The tubes were incubated in 37oC for 18-24 hours and the turbidity was inspected. The results were planted on NAP, incubated, and the growth of colonies was inspected. The research was repeated four times. From the inspection, it is found that the Minimal Inhibitory Concentration (MIC) was 13,5% and the Minimal Bactericidal Concentration (MBC) was 15%. Using Kruskal-Wallis, a significancy value of p<0,05 was found. In conclusion, weeping fig (Ficus benjamina L.) leaves extract has antibacterial effect towards E. coli in vitro.
