-Methods: Angiotensin II was treated to the vascular smooth muscle cells (VSMCs) to stimulate the circadian oscillation of clock and clock-controlled genes in VSMCs. Suppressive subtractive hybridization method was used to obtain differentially expressed gene in circadian manner. Finally, northern blot were performed to confirm the circadian expression in mice aorta. The mice were kept in 12;12 h light-dark cycle. Results: We observed circadian oscillation of gene expression of Matrix Metalloproteinase-9 (MMP-9) and LDL Receptor Related Protein-1 (LRP-1), the genes involved in extracellular matrix degradation, in Angiotensin II treated VSMC. Furthermore the cycling of MMP-9 and LRP-1 also detected mice aorta. In the heart, Matrix Metalloproteinase-2 (MMP-2), Tissue Inhibitor of MMP 1 (TIMP-1) and LRP-1 mRNA also possess circadian rhythm Therefore, circadian expression of MMP-9, LRP-1 MMP-2 and TIMP-1 may contribute to the circadian pattern of cardiovascular events.
