Abstract

Latar Belakang: Transforming Growth Factor Beta (TGF-β) menurun pada mencit dengan Lupus Eritematosus Sistemik (LES). Pengembangan metode elicit dose immunotherapy menggunakan self antigen dsDNA menginduksi toleransi sistem imun sehingga berpotensi dalam regulasi sistem imun. Tujuan: Penelitian ini dilaksanakan untujk mengetahui efek pemberian berbagai dosis self antigen dsDNA secara bertahap terhadap kadar TGF-β pada mencit model LES yang diinduksi pristane. Metode: Penelitan ini merupakan eksperimental murni (true experimental) laboratorik dengan desain post test only controlled group design menggunakan mencit Balb/c sebagai subjek diinjeksi pristane dan setelah itu self antigen dsDNA berbagai dosis secara berurutan dan bertahap. Hasil: Hasil dari penelitian ini didapatkan bahwa pengaruh pemberian self antigen dsDNA berbagai dosis terhadap kadar TGF-β. Kelompok kontrol positif (261.02 ± 52.26 pg/ml), Kontrol negatif (335.38 ± 27.81 pg/ml) perlakuan A (732.43 ± 98.69 pg/ml), perlakuan B (740.90 ± 253.41 pg/ml) dan perlakuan C (382.25 ± 96.13 pg/ml). Diketahui perlakuan A dan B berbeda signifikan dengan kelompok kontrol positif. Kesimpulan: Data penelitian menunjukkan peningkatan dosis self antigen dsDNA mempengaruhi kadar TGF-β terutama pada dosis dengan konsentrasi 0.1 μg/ml (0.5 ml), 1 μg/ml (0.5 ml), 10 μg/ml (0.5 ml) sehingga mampu memperbaiki regulasi sistem imun dan menimbulkan terjadinya toleransi dan desensitisasi sistem imun.

English Abstract

Background: Transforming Growth Factor (TGF-β) decreased in patients with Systemic Lupus Erythematosus (SLE). Elicit dose immutoherapy method using self antigen developed in induced immune tolerance system in order to improvise the regulation of the immune system in animal model. Objective: Study was conducted to know the effects of multiple doses of self antigen dsDNA to the amount of TGF-β in pristane-induced SLE on animal model. Method: Study used a true experimental laboratory design with post test only controlled group design used Balb/c mice as the subject of injected pristane and multiple doses of self antigen dsDNA gived gradually and sequentially. Results: The results showed the effects of administered of self antigen dsDNA at multiple doses to the amount of TGF-β. From the group of negative control (335.38 ± 27.81 pg / ml), positive control 261.02 ± 52.26 pg / ml), group A (732.43 ± 98.69 pg / ml), group B (740.90 ± 253.41 pg / ml) and group C (382.25 ± 96.13 pg / ml). The value of TGF-β was significantly different in group A and B compare with positive control. Conclusion: Research data showed an increase doses of self antigen dsDNA influence the amount of TGF-β especially at dose with concentration 0.1 μg / ml (0.5 ml), 1 μg / ml ( 0.5 ml), 10 μg / ml (0.5 ml) in improve immune system regulation and lead to tolerance and desensitization of the immune system.
