Abstract
Latar Belakang: Hipertensif Portal Gastropati (HPG) adalah salah satu komplikasi fibrosis hati akibat hipertensi portal, kerusakan oksidatif berperan didalamnya. MDA merupakan produk akhir dari peroksidasi lipid pada membran sel, menggambarkan kerusakan jaringan oksidatif. GPx adalah enzim antioksidan yang berperan menjaga integritas jaringan dari kerusakan oksidatif. Pemberian kurkumin sebagai antioksidan diharapkan dapat memperbaiki HPG. Tujuan: Mengetahui hubungan antara pemberian kurkumin terhadap reaksi stres oksidatif dan gambaran histopatologi lambung pada tikus model fibrosis. Metode: Penelitian eksperimental pada tikus jantan strain wistar. Terdapat 8 kelompok, masing-masing terdiri dari 4 tikus. Kelompok KP dipapar CCl4 1cc/kgbb 2x perminggu intraperitoneal selama 9 minggu untuk membentuk fibrosis F3 dan HPG. Kelompok KN diinjeksi Normal Salin. Setelah diinjeksi CCl4, Kelompok Kontrol (KK) diberikan plasebo dan Kelompok Perlakuan (KP2, KP5 dan KP9) diberikan kurkumin 200mg/kgbb 2, 5 dan 9 minggu. Dilakukan analisis kadar MDA, aktivitas GPx dan histopatologi lambung dengan menggunakan uji t independen, uji pengaruh dan lama pemberian kurkumin dengan ANOVA. Hasil: Terdapat peningkatan signifikan kadar MDA dan skor histopatologi lambung pada kelompok KP, sedangkan aktivitas GPx menurun nyata dibandingkan kelompok KN, Kadar MDA menurun pada pemberian kurkumin, menurun nyata pada KP5 dan KP9. GPx meningkat pada pemberian kurkumin, meningkat nyata pada KP5. Gambaran histopatologi mengalami perbaikan pada KP5, memburuk pada KP9, kemungkinan karena faktor lain yang memicu cedera lambung. Kesimpulan: Terdapat penurunan signifikan kadar MDA jaringan lambung, peningkatan signifikan aktivitas GPx setelah pemberian kurkumin, sedangkan gambaran histopatologi lambung tidak terdapat perbaikan yang signifikan. Lama pemberian kurkumin berkorelasi dengan penurunan MDA dan penurunan GPx, tetapi tidak berkorelasi dengan histopatologi lambung
English Abstract
Background: Portal Hypertensive Gastropathy (PHG) is one of the complications of liver cirrhosis due to portal hypertension, oxidative damage plays a role in it. MDA is the final product of lipid peroxidation in cell membranes, representing oxidative tissue damage. GPx is an antioxidant enzyme maintaining tissue integrity from oxidative damage. Curcumin administration as an antioxidant is expected to improve PHG. Objective: To determine the effects of curcumin against oxidative stress reactions and the overview of gastric histopathology. Methods: An experimental study in male rats wistar strain. There are 8 groups, 4 rats for each. KP group was exposed to CCl4 1cc / kg intraperitoneally 2 times per week for 9 weeks to form F3 fibrosis and PHG. KN group was injected Normal Saline. Once injected CCl 4 Control Group (KK) was given a placebo and intervention groups (KP) was given curcumin 200mg per kg weight for 2, 5 and 9 weeks. Analysis of MDA level, GPx activity gastric tissue and histopathology is conducted by independent t-test, influence test and the duration of administration curcumin by ANOVA. Result: MDA levels of tissue and gastric histopathology-based score in the KP group remarkably increased, whereas the GPx of the tissue is decreased significantly compared to KN group. Curcumin decreases MDA level as shown in KP5 and KP9 that the MDA levels are significantly decreased. Curcumin increases GPx levels as shown in in KP 5 that the GPx levels are significantly increased. Histopathology improved on KP5, worse in KP9, probably due to other factors that trigger gastric injury. Conclusion: The MDA levels of gastric tissue are decreased, GPx activity are increased and gastric histopathology are improved by the administration of curcumin. The duration of curcumin giving is correlated with the decreased of MDA level and GPx activity, but is not correlated with gastric histopathology.
