Abstract
Nikotin adalah zat kimia yang banyak dihasilkan dari tanaman tembakau atau diproduksi secara sintetis. Dalam penelitian invitro nikotin dapat meningkatkan aktivitas osteoblas dan menurunkan aktivitas osteoklas. Pada wanita menopause kadar estrogen cenderung menurun sehingga beresiko mengalami osteoporosis. Osteoporosis sendiri terjadi karena ketidakseimbangan aktivitas osteoblas dan osteoklas. Penelitian ini dilakukan untuk melihat pengaruh pemberian larutan nikotin terhadap kadar deoxypiridinoline (DPD) serum sebagai marker resorpsi tulang pada tikus wistar betina (Rattus novergicus) model osteoporosis. Penelitian ini adalah penelitian true experimental (eksperimental murni) di laboratorium secara in vivo. Penelitian dilakukan dengan desain post test only pada kelompok perlakuan dan kontrol. Pengelompokan objek eksperimen dilakukan dengan desain randomisasi sederhana. Tikus wistar yang digunakan berjumlah 25 ekor dan dibagi menjadi 5 kelompok, kelompok 1 sebagai kontrol negatif, kelompok kedua sebagai kontrol positif yang hanya dilakukan bilateral oovorektomi dan 3 kelompok selanjutnya sebagai kelompok perlakuan juga dilakukan bilateral oovorektomi serta pemberian dosis nikotin yang berbeda. Kelompok perlakuan 1 diberi dosis nikotin sebesar 0,25 mg/kgBB, kelompok perlakuan 2 dengan dosis nikotin 0,5 mg/kgBB dan kelompok perlakuan 3 dosis nikotin 0,75mg/kgBB. Pemberian nikotin dilakukan selama 5 minggu. Hasil penelitian menunjukkan penurunan kadar DPD serum sebesar 19,2% pada uji regresi linier dan tidak memiliki perbedaan yang bermakna pada tiap kelompok perlakuan (Post Hoc Tukey HSD, p > 0,05). Oleh karena itu dapat disimpulkan bahwa pemberian larutan nikotin terhadap kadar DPD serum tidak memiliki pengaruh yang signifikan pada tikus model osteoporosis.
English Abstract
Nicotine is a chemical substance that produced by tobacco or synthectically. In an invitro research nicotine is able to increase osteoblasts activity and decrease osteoclasts activity. In postmenopausal women, the level of estrogen tends to be decreased, giving risk of developing osteoporosis. Osteoporosis itself occurs because of the imbalance between osteoblasts and osteoclasts activity. This research have been done to look for the effect of nicotine solution on levels of serum deoxypiridinoline (DPD) as a marker of bone resorbtion in osteoporotic Rattus novergicus. This true experimental research was conducted in laboratory in vivo and was using post test only design with experimental and control groups. The experimental groups’s subjects were randomized using simple randomization design. The amount of mice that was used in this research are 25 mice, divided into 5 groups; first group as negative control, second group as positive control, that only bilateral oovorectomied and the rest of the group are treatment groups that also oovariectomied and given different doses of nicotine. The first treatment group are given the nicotine 0.25 mg/kgBB, the second treatment group are given the nicotine 0.5 mg/kgBB and the third treatment group are given nicotine 0.75 mg/kgBB. The nicotine was given for 5 weeks. The result of the research showing decreasing DPD serum levels as much as 19.2% on linear regression test and have no significant differences on each treatment group (Post Hoc Tukey HSD, p >0.05). So it can be concluded that the nicotine solution effect on serum DPD levels has no significant effect on osteoporotic mice.
