Abstract

Gastric ulcer is a type of peptic ulcer that occurs in the lining of the stomach. Gastric ulcer involves an imbalance between aggressive factors, such as gastric acid secretion, pepsin, and Helicobacter pylori infection, and protective factors, such as mucus secretion, bicarbonate secretion, and blood flow to the stomach lining. The management of gastric ulcers involves a combination of pharmacological therapy and lifestyle modifications. Proton pump inhibitors (PPIs), such as omeprazole, esomeprazole, and pantoprazole, are the first-line treatment for gastric ulcers as they effectively reduce gastric acid secretion and promote ulcer healing. it is necessary to explore other natural ingredients that have antinflammatory properties, one of which is the plant Rosmarinus officinalis L. Nanomaterials have been increasingly used in drug delivery systems due to their unique physicochemical properties, such as high surface area, small size, and surface reactivity; So in this research, nanoparticles were used in the hope of providing a better pharmacokinetic effect. This study used a true experimental design - randomized posttest only controlled group design in vivo. There was a significant difference in the expression of TNF-α in the negative control group and the positive control group. This proves that stomach ulcers are produced using 2 ml/kg body weight of 96% ethanol. There was a decrease in the expression of IL-6 and TNF-α in the nanoemulsion treatment group with 25 mg/mL RO extract; 50mg/ml; and 100mg/ml. The effective concentration in reducing the expression of IL-6 and TNF- α occurred at a concentration of 100 mg/mL. Therefore, it can be concluded that administration of Rosmarinus officinalis L nanoemulsion can inhibit IL-6 and TNF-α in mice models of gastric ulcer.

English Abstract

Tukak lambung merupakan salah satu jenis tukak lambung yang terjadi pada lapisan lambung. Tukak lambung melibatkan ketidakseimbangan antara faktor agresif, seperti sekresi asam lambung, pepsin, dan infeksi Helicobacter pylori, dengan faktor protektif, seperti sekresi mukus, sekresi bikarbonat, dan aliran darah ke lapisan lambung. Penatalaksanaan tukak lambung melibatkan kombinasi terapi farmakologis dan perubahan gaya hidup. Penghambat pompa proton (PPI), seperti omeprazole, esomeprazole, dan pantoprazole, merupakan pengobatan lini pertama untuk tukak lambung karena obat-obatan tersebut efektif mengurangi sekresi asam lambung dan mempercepat penyembuhan tukak. Perlu dikaji bahan-bahan alami lain yang memiliki sifat antiinflamasi, salah satunya adalah tanaman Rosmarinus officinalis L. Nanomaterial semakin banyak digunakan dalam sistem penghantaran obat karena sifat fisikokimianya yang unik, seperti luas permukaan yang tinggi, ukuran yang kecil, dan reaktivitas permukaan; Maka pada penelitian ini digunakan nanopartikel dengan harapan dapat memberikan efek farmakokinetik yang lebih baik. Penelitian ini menggunakan true experimental design - randomized posttest only controlled group design in vivo. Terdapat perbedaan ekspresi TNF-α yang signifikan pada kelompok kontrol negatif dan kelompok kontrol positif. Hal ini membuktikan bahwa tukak lambung terbentuk dengan menggunakan etanol 96% sebanyak 2 ml/kg berat badan. Terjadi penurunan ekspresi IL-6 dan TNF-α pada kelompok perlakuan nanoemulsi dengan ekstrak RO 25 mg/mL; 50 mg/ml; dan 100 mg/ml. Konsentrasi yang efektif dalam menurunkan ekspresi IL-6 dan TNF-α terjadi pada konsentrasi 100 mg/mL. Dengan demikian dapat disimpulkan bahwa pemberian nanoemulsi Rosmarinus officinalis L dapat menghambat IL-6 dan TNF-α pada mencit model tukak lambung.
