Abstract

Tujuan penelitian ini adalah untuk menganalisis kandungan total flavonoid, antioksidan, dan aktivitas antioksidan pada daun beberapa nilam tetraploid hasil induksi in vitro. Langkah penelitian ini terdiri atas beberapa tahap, yaitu ekstraksi daun nilam, analisis kandungan total flavonoid dan antioksidan, dan uji aktivitas antioksidan ekstrak daun nilam. Daun nilam diploid dan tetraploid T1, T2, T3, T4, T5, T7 diekstraksi dengan metanol untuk didapatkan ekstrak cair nilam. Uji kandungan total flavonoid, kandungan total antioksidan, & aktivitas antioksidan ekstrak nilam dilakukan dengan metode AlCl3, fosfomolybdenum, & DPPH. Tingkat aktivitas antioksidan ditentukan berdasarkan IC50. Rancangan penelitian ini rancangan acak lengkap dengan tiga kali ulangan. Analisis data menggunakan one-way ANOVA untuk perbandingan & nilai R2 pada analisis regresi untuk korelasi. Kandungan total flavonoid nilam tetraploid bervariasi (0,52 � 0,64 mg QE) di mana tetraploid T1, T4, & T7 lebih tinggi dan tetraploid T2, T3, & T5 cenderung lebih tinggi dibandingkan nilam diploid. Kandungan total antioksidan nilam tetraploid juga bervariasi (2,81 � 3,67 mg QE) di mana tetraploid T1, T5, & T7 cenderung lebih tinggi, sedangkan tetraploid T2, T3, & T4 lebih rendah dibandingkan nilam diploid. Aktivitas antioksidan nilam diploid dan semua tetraploid sangat kuat, namun terdapat perbedaan nilai IC50. Nilai IC50 nilam tetraploid T1 & T4 lebih rendah dibandingkan nilam diploid yang mengindikasikan aktivitas antioksidan yang lebih baik. Kandungan total flavonoid dan antioksidan nilam tetraploid berkorelasi lemah dengan aktivitas antioksidan yang mengindikasikan terdapat beberapa senyawa fitokimia lain yang memiliki pengaruh terhadap aktivitas antioksidan.

English Abstract

Goals of this research were to analyse total flavonoid and antioxidant content, and antioxidant activity in the leaves of several in vitro-induced tetraploid patchouli types. Method of this research consisted of several steps, which are patchouli leaves extraction, total flavonoid and antioxidant content analysis, and antioxidant activity assay. Diploid and tetraploid T1, T2, T3, T4, T5, T7 patchouli leaves were extracted with methanol to obtain crude patchouli leaves extract. Total flavonoid content, total antioxidant content, and antioxidant activity assay of patchouli extract was done with AlCl3, phosphomolybdenum, & DPPH method. Antioxidant activity level was determined based on IC50. Research design for this study was complete random design with three replications. Data analysis was done by one-way ANOVA for comparison and R 2 value in regression analysis for correlation. Total flavonoid content of tetraploid patchouli was varied (0,52 – 0,64 mg QE) where tetraploid T1, T4, & T7 were higher and tetraploid T2, T3, & T5 were tended to be higher than diploid type. Total antioxidant content of tetraploid patchouli was also varied (2,81 – 3,67 mg QE) where tetraploid T1, T5, & T7 were tended to be higher, whereas tetraploid T2, T3, & T4 were lower than diploid type. Antioxidant activity of diploid and all tetraploid patchouli were very strong, but IC50 values were different. IC50 value of tetraploid patchouli T1 & T4 were lower than diploid type, indicating better antioxidant activity. Total flavonoid and antioxidant content of tetraploid patchouli had weak correlation with antioxidant activity, indicating other phytochemical compounds have influence for antioxidant activity.
