Abstract
Eucheuma cottonii merupakan jenis alga merah yang mudah ditemukan di perairan Asia Tenggara. Berdasarkan beberapa penelitian, Eucheuma cottonii dari beberapa sumber perairan memiliki keragaman kandungan senyawa bioaktif dan pigmen fotosintetik disebabkan oleh lokasi budidaya, praktek budidaya dan kondisi perairanyang berbeda. Senyawa bioaktif memberikan pengaruh terhadap aktivitas antioksidan dan antibakteri. Aktivitas farmakologi ini dapat dimanfaatkan sebagai alternatif penanganan penyakit degeneratif dan foodborne disease yang merupakan penyumbang data mortalitas terbesar di Indonesia bahkan Asia. Informasi mengenai aktivitas Sedikitnya informasi mengenai antioksidan dan antibakteri dari ekstrak Eucheuma cottonii masih kurang. Penelitian ini bertujuan mengetahui pengaruh metode Microwave Assisted Extraction terhadap aktivitas antioksidan dan antibakteri dari ekstrak Eucheuma cottonii dari perairan Probolinggo, Jawa Timur. Penelitian ini merupakan penelitian eksperimental skala laboratorium dengan rancangan percobaan acak kelompok (RAK) dengan 2 faktor, yaitu: suhu ekstraksi (50, 60, 70°C) dan waktu ekstraksi (10, 20, 30 menit) dengan dilakukan 3 ulangan. Aktivitas antioksidan diukur dengan metode pengikat radikal bebas 2,2-diphenyl-1- picrylhydrazyl (DPPH). Aktivitas antibakteri diukur dengan metode Well Diffusion. Analisa GC-MS digunakan untuk mengidentifikasi asam lemak. Analisa LC-MS digunakan untuk mengidentifikasi senyawa flavonoid. Analisa data menggunakan analisa ragam (ANOVA) dan dilanjutkan dengan Duncan Multiple Range Test (DMRT). Perlakuan terbaik ditentukan dengan metode Multiple Atribut (Zeleny). Hasil penelitian menunjukkan persentase rendemen terus meningkat dengan semakin bertambahnya suhu dan waktu ekstraksi. Nilai hasil uji total fenol dan flavonoid terus meningkat sampai perlakuan suhu ekstraksi 60°C dan waktu ekstraksi 10 menit kemudian terjadi penurunan dengan bertambahnya suhu dan waktu ekstraksi. Sama halnya dengan nilai IC50, nilai terus menurun sampai perlakuan suhu ekstraksi 60°C dan waktu ekstraksi 10 menit kemudian terjadi peningkatan dengan bertambahnya suhu dan waktu ekstraksi. Hasil uji antibakteri terhadap kedua bakteri menunjukkan data fluktuatif, zona terluas pada perlakuan suhu ekstraksi 60°C dan waktu ekstraksi 10 menit sedangkan zona terkecil pada perlakuan suhu ekstraksi 70°C dan waktu ekstraksi 30 menit. Perlakuan terbaik diperoleh kombinasi perlakuan suhu ekstraksi 60°C dan waktu ekstraksi 10 menit dengan karakteristik rendemen 2,71%, total fenol 103,94 mg GAE/g, total flavonoid 302,97 mg EQ/g, nilai IC50 305,87 ppm, diameter zona bening bakteri Escherichia coli 14,84 mm dan diameter zona bening bakteri Staphylococcus aureus 16,64 mm. Analisa LC-MS mengidentifikasi epigenin, rutin dan kuersetin sebagai senyawa flavonoid. GC-MS mengidentifikasi asam lemak yaitu n-hexadecanoic acid, octadecanoic acid, dan 9-octadecenoic acid.
English Abstract
Eucheuma cottonii is a one of red algae species commonly found in Southeast Asia’s coastal area. Based on previous studies, Eucheuma cottonii from different coastal sources contain diverse bioactive compounds and photosynthetic pigments caused by the cultivation location, cultivation practices and differences in water conditions. Bioactive compounds give effect for antioxidant and antibacterial activity. This pharmacological activity can be used as an alternative treatment of degenerative diseases and foodborne diseases which is the largest contributor of mortality rate in Indonesia and even Asia. Nonetheless, information about antioxidant and antibacterial activity of Eucheuma cottonii extract is limited. The aim of the present study was to investigate the effect of Microwave Assisted Extraction method on antioxidant and antibacterial activity of Eucheuma cottonii extract from Probolinggo coast of East Java. The research was a laboratory experimental study with randomized block design consisted of 2 factors, namely: extraction temperature (50, 60, 70°C) and extraction time (10, 20, 30 min) with 3 replications. The antioxidant activity was measured by 2,2- diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay. The antibacterial activity was measured by Well Diffusion method. GC-MS was performed for the identification of fatty acids. LC-MS was performed for the identification of flavonoid compounds. Data were analyzed using analysis of variance (ANOVA) and continued with Duncan Multiple Range Test (DMRT). The best treatment was determined by using Multiple Attribute (Zeleny) method. The result showed that the longer extraction time and the higher extraction temperature increased the extraction yield. Total phenol and flavonoid was increased until the temperature of 60°C for 10 minutes, then decreased with longer time and higher temperature of extraction. IC50 value was getting lower until the temperature of 60°C for 10 minutes, then increased IC50 value was observed with higher temperature and longer time, suggesting decreased antioxidant activity. Antibacterial activity showed similar result by means of clear zone for Escherichia coli and Staphylococcus aureus. The biggest clear zone for both strains was produced by extract yielded from extraction temperature of 60°C for 10 minutes and the smallest clear zone was produced by extract yielded from extraction temperature of 70°C for 30 minutes. The result of the present study showed that the best treatment was obtained by a combination of extraction temperature of 60°C and extraction time of 10 minutes which was characterized by 2.71% of yield, 103.94 mg GAE/g of total phenolic content (TPC), 302.97 mg EQ/g of total flavonoid content (TFC), 306.87 ppm of IC50 value, 14.84 mm of clear zone inhibition of Escherichia coli, and 16.64 mm of clear zone inhibition of Staphylococcus aureus. LC-MS analysis identified Apigenin, rutin, and quercetin among flavonoid compounds in Eucheuma cottonnii. GC-MS analysis identified n-hexadecanoic acid, octadecanoic acid, and 9-octadecenoic acid among fatty acids in Eucheuma cottonnii.
