Abstract

Salmonelosis dapat disebabkan oleh S.enterica. Gejala klinis yang dapat teramati yaitu diare. S.enterica dapat memicu terjadinya kerusakan pada ileum. Kandungan kopi robusta Lampung seperti kafein, CGA dan alkaloid dapat sebagai antibakteri sehingga diharapkan dapat menekan jumlah S.entericadan dikombinasikan dengan L. acidophilus sebagai kompetitor S.enterica. Tujuan dari penelitian ini yaitu mengetahui pengaruh pemberian kombinasi ekstrak kopi robusta Lampung dan Lactobacillus acidophilus terhadap jumlah bakteri pada ileum dan perbaikan organ ileum pada mecit Balb/C model salmonellosis. Rancangan penelitian yang digunakan dalam penelitian ini yaitu studi eksperimental dengan metode RAL dengan pengambilan data post test only control design. Hewan coba menggunakan mencit Balb/C jantan umur 8 minggu, BB20-25 gram sebanyak 24 ekor. Terdapat 6 kelompok perlakuan yaitu : K- (sehat), K+ (S. enterica), K. Lactobacillus (L. acidophilus dan S. enterica), P1,P2 dan P3 merupakan kelompok yang diberikan kombinasi ekstrak kopi robusta Lampung dan L. acidophilus. Dosis ekstrak kopi robusta Lampung yang digunakan yaitu mulai dari 250 mg/kg BB, 500 mg/kg BB dan 750 mg/kg BB. Parameter yang diamati yaitu jumlah bakteri pada ileum menggunakan metode TPC. Gambaran histopatologi ileum dianalisis secara deskriptif. Berdasarkan hasil yang didapat, pemberian kombinasi ekstrak kopi robusta Lampung dengan dosis 250 mg/kg BB dan L. acidophilus dapat meningkatkan jumlah bakteri serta mencegah kerusakan ileum.

English Abstract

Salmonelosis is a disease that can be caused by Salmonella enterica. Clinical signs that can be observed are gastroenteritis and diarrhea. S. enterditis entered the body and initiate damage towards the ileum. Contents from Lampung Robusta coffee such as caffeine, CGA and alkaloid has antibacterial properties which can be used to decrease the amount of infection caused by S. enterica combined with Lactobacillus acidophilus as a competitor. The aim of this research is to acknowledge the effect of combination of Lampung Robusta coffee extract with L. acidophilus towards the quantity of bacteriaand the ileum's healing in salmonelosisBalb/C mice. This research used experimental study with RAL method which takes data in post test only control design. The 24 mice used were all Balb/C male mice, aged 8-10 weeks with body weight around 20-25 grams. The mice was then divided into 6 groups with different treatments. First group was used as negative controls with no induction, and the second was used as the positive control which was induced by S. enterica. The third group was induced by S. entericaand was given a shot of L. acidophilus. Group 4 (P1), group 5 (P2) and group 6 (P3) were all induced by S. entericaand combination of Lampung Robusta coffee extract with L. acidophilus. Dosage of the Lampung Robusta coffee extract used in this research was 250 mg/Kg (P1), 500 mg/Kg (P2) and 750 mg/Kg (P3). Parameters studied in this research are the quantity of bacteria using total plate count method, and descriptive analysis using histopathology of the ileum.Based on the results obtained, the combination of Lampung Robusta coffee extract with a dose of 250 mg /Kg and L. acidophilus can increase the quantity of bacteria and prevent ileum damage.
