Abstract

Tujuan: Mengetahui efek pemberian jus kecambah brokoli dalam mencegah agregasi protein lensa pada tikus model diabetes. Metode: Penelitian ini merupakan eksperimental laboratoris in vivo pada 25 tikus Wistar jantan yang dibagi dalam kelompok kontrol negatif, tikus diabetes tanpa perlakuan jus sebagai kontrol positif dan tikus diabetes yang diberi jus kecambah brokoli 0,25 gram/hari, 0,5 gram/hari dan 1 gram/hari selama 28 hari. Kondisi diabetes didapatkan dengan pemberian streptozotocin 65 mg/kgbb single dose intraperitoneal. Pada akhir perlakuan, semua tikus dilakukan pemeriksaan kadar gula darah puasa (GDP), derajat katarak menggunakan biomikroskopi slit lamp dan ekspresi αB-crystallin lensa menggunakan metode Western blot. Hasil: Perlakuan jus kecambah brokoli secara signifikan menurunkan derajat katarak, kadar glukosa darah, dan ekspresi αB-crystallin lensa agregrasi, namun meningkatkan ekspresi αB-crystallin native lensa pada tikus model diabetes (p <0,05). Kesimpulan: Jus kecambah brokoli berpengaruh yang signifikan dalam mencegah agregasi protein lensa pada tikus model diabetes, dimana semakin tinggi dosis yang diberikan akan semakin memberikan gambaran derajat katarak yang lebih ringan, ekspresi αB-crystallin native lensa yang lebih tinggi dan ekspresi αB-crystallin agregrasi lensa yang lebih rendah.

English Abstract

AIM: To investigate the effect of broccoli sprouts juice in preventing lens protein aggregation in diabetic rat model. METHODS: This study was a laboratory experimental in vivo of 25 male Wistar rats. They were divided into negative control group, diabetic group without juice treatment as positive control and diabetic group given broccoli sprouts juice 0.25 g/day, 0.5 g/day and 1 g/day for 28 days. Diabetic rat model was obtained by administering a single dose streptozotocin 65 mg/kg intraperitoneal. At the end of the study, all rats were examined for fasting blood sugar level (FBS), visual cataract score using slit lamp biomicroscope and lens αB-crystallin expression using Western blot method. And statistical analysis was performed using one way ANOVA with post hoc test, correlation test, and simple linear regression. RESULTS: Broccoli sprout juice treatment significantly reduced cataract scores, blood glucose levels, and lens aggregated αB-crystallin expression, but increased the expression of lens native αB-crystallin in diabetic rat models (p<0.05). CONCLUSION: Broccoli sprout juice has a significant effect in preventing lens protein aggregation in diabetic rat model. The higher dose gives better visual cataract scores, lower lens aggregated αB-crystallin expression and higher lens native αB-crystallin expression.
