Abstract
Hiperkolesterolemia merupakan suatu keadaan meningkatnya kadar kolesterol di dalam darah melebihi batas normal terjadi akibat akumulasi kolesterol dan lipid pada dinding pembuluh darah. Senyawa yang terdapat pada jamur kuping hitam yaitu tanin, polisakarida, dan niasin. Kandungan tersebut dapat menurunkan kadar trigliserida dengan meningkatkan aktivitas enzim LPL. Tujuan penelitian ini adalah untuk mengetahui pengaruh pemberian air rebusan jamur kuping hitam (Auricularia polytricha) untuk menurunkan kadar trigliserida dan memperbaiki kerusakan gambaran histopatologi aorta pada tikus putih (Rattus norvegicus) model hiperkolesterolemia. Penelitian ini menggunakan Rancangan Acak Lengkap yang terdiri dari lima kelompok, yaitu kontrol negatif, kontrol positif (diet hiperkolesterol selama 42 hari), dan tiga kelompok perlakuan dengan diberi diet hiperkolesterol selama 42 hari dan diberi terapi air rebusan jamur kuping hitam secara per oral selama 14 hari dengan dosis 250 mg/kgBB, 500 mg/kgBB, dan 750 mg/kgBB. Pengukuran kadar trigliserida dengan menggunakan Spektrofotometer Biosystem A15. Pengamatan histopatologi aorta dilakukan dengan pewarnaan HE. Analisis data menggunakan Uji ANOVA menggunakan SPSS 22. Pengamatan histopatologi aorta secara deskriptif kualitatif. Hasil penelitian menunjukkan terapi air rebusan jamur kuping hitam dengan dosis 250 mg/kgBB tidak berbeda nyata (p>0,05) dengan kelompok tikus sehat. Dosis terapi 250 mg/kgBB merupakan dosis terbaik karena mampu memperbaiki tunika intima dan tunika media. Disimpulkan bahwa air rebusan jamur kuping hitam dapat digunakan sebagai terapi hiperkolesterolemia berdasarkan penurunan kadar trigliserida dan memperbaiki kerusakan pada tunika intima dan tunika media.
English Abstract
Hypercholesterolemia is a condition of increasing cholesterol levels in the blood beyond normal limits due to the accumulation of cholesterol and lipids in the blood vessel walls. Black ear mushroom contains tannins, polysaccharide, and niacin which can reduce triglyceride levels by increasing LPL enzyme. The purpose of this study was to determine the effect of black ear mushroom boiled water (Auricularia polytricha) to reduce the levels of triglycerides and repair damage to the aortic histopathology in the hypercholesterolemic rat (Rattus norvegicus) model. This study ia a experimental laboratory using a Completely Randomized Design consisting of five groups, namely negative control, positive control (received a hypercholesterolemia diet for 42 days), and three groups with a hypercholesterolemia diet for 42 days and treated with of black mushroom cooking water with oral therapy for 14 days with a dose of 250 mg/kg body weight, 500 mg/kg body weight, and 750 mg/kg body). Triglyceride levels was measured by using the Biosystem A15 Spectophotometer. Observation of aortic histopathology was carried out by staining Hematoxylin Eosin (HE). Data analysis used the ANOVA test using SPSS 22. Observation of aortic histopathology was in a descriptive manner. The results showed that the treatment of black mushroom boiled water at a dose of 250 mg/kg body weight does not significantly different (p>0,05) from the group of healthy rat. The therapeutic dose of 250 mg/kg body weight is the best dose that can improve tunica intima and tunica media. It was concluded that black mushroom decoction water containing tanin compounds could be used as a therapy for hypercholesterolemia based on a decrease in triglyceride levels and improved tunica intima and tunica media.
