Abstract

Nekrosis pulpa merupakan kematian jaringan pulpa karena sistem pertahanan pada pulpa tidak lagi dapat menahan besar rangsangan. Perawatannya nekrosis pulpa adalah perawatan saluran akar. Salah satu faktor yang mempengaruhi hasil perawatan saluran akar adalah adanya bakteri pada saluran akar, salah satunya yaitu Streptococcus sanguinis. Ekstrak rimpang kencur mengandung senyawa aktif antara lain alkaloid, flavonoid, polifenol, saponin, dan tanin. Penelitian ini bertujuan membuktikan ekstrak rimpang kencur efektif sebagai antibakteri Streptococcus sanguinis secara in vitro. Penelitian ini bersifat true experimental dengan metode difusi sumuran. Ekstraksi rimpang kencur menggunakan metode maserasi. Konsentrasi ekstrak rimpang kencur yang digunakan adalah 20%, 40%, 60%, 80%, 100%, kontrol negatif aquades dan kontrol positif kalsium hidroksida. Hasil penelitian menunjukkan ekstrak rimpang kencur efektif sebagai antibakteri Streptococcus sanguinis dimulai pada konsentrasi 40% ditandai terbentuknya zona hambat diameter 9,2 mm. Diameter zona hambat semakin besar seiring besarnya konsentrasi ekstrak rimpang kencur, sedangkan zona hambat pada konsentrasi 100% mempunyai diameter yang sama dengan kalsium hidroksida. Analisis data menggunakan One-way ANOVA menunjukkan perbedaan yang signifikan pada perubahan konsentrasi ekstrak rimpang kencur terhadap zona hambat Streptococcus sanguinis (p<0,05). Berdasarkan penelitian ini, dapat disimpulkan bahwa ekstrak rimpang kencur (Kaempferia galangal L.) mempunyai efek antibakteri terhadap Streptococcus sanguinis secara in-vitro.

English Abstract

Necrotic pulp is the death of pulp tissue due to the pulp defense system which can no longer resist the amount of stimulation. The treatment is root canal treatment. One of the factor that can affect the results of root canal treatment is the presence of bacteria in the root canal, one of the bacteria in the root canal is Streptococcus sanguinis. Kencur rhizome extract contains active chemical compounds including alkaloids, flavonoids, polyphenols, saponins, and tannins. This research aims to prove that the kencur rhizome extract is effective as an antibacterial for Streptococcus sanguinis with in vitro test. This research is true experimental with well diffusion method. Kencur rhizome extract is made by maceration method. The concentration of kencur rhizome extract used was 20%, 40%, 60%, 80%, 100%, with aquades as negative control, and calcium hydroxide as positive control. The results of this research showed that kencur rhizome extract is effective as an antibacterial of Streptococcus sanguinis began at a concentration of 40% proven by inhibition zone diameter 9,2 mm. The diameter of the inhibition zone increases along with the amount of the concentration of kencur rhizome extract, while the inhibition zone at a 100% concentration has the same diameter as calcium hydroxide. Data analysis using One-way ANOVA showed a significant difference in changes in the concentration of kencur rhizome extract against the Streptococcus sanguinis inhibition zone (p<0,05). Based on this research, it can be concluded that the extract of kencur rhizome (Kaempferia galangal L.) has an antibacterial effect on Streptococcus sanguinis with in vitro.
