Abstract

Anak dengan Autism Spectrum Disorder (ASD) mengalami kondisi dysbiosis atau ketidakseimbangan jumlah mikroflora usus yang mengakibatkan jumlah bakteri menguntungkan (probiotik) lebih rendah daripada bakteri merugikan (patogen). Prebiotik adalah bahan pangan yang tidak mampu dicerna oleh tubuh namun membawa manfaat dengan menstimulasi pertumbuhan bakteri probiotik. Lactobacillus acidophilus dan Lactobacillus plantarum tergolong Bakteri Asam Laktat (BAL) yang cukup sering dimanfaatkan sebagai bakteri probiotik. Bahan pangan yang memiliki kandungan prebiotik salah satunya adalah kacang hijau dan pisang, sehingga hasil olahannya dapat digunakan untuk meningkatkan pertumbuhan bakteri probiotik. Penelitian ini termasuk experimental design dengan 2 tahapan yaitu penelitian pendahuluan untuk mengetahui kandungan pati resisten cookies prebiotik dan penelitian utama yaitu pengujian pertumbuhan bakteri probiotik Lactobacillus acidophilus dan Lactobacillus plantarum untuk melihat efektifitas pemberian cookies prebiotik yang dilakukan dengan 2 perlakuan dan 2 pengulangan untuk masing-masing bakteri. Hasil dari analisis statistik menggunakan Independet Sample T-test menunjukkan bahwa terdapat perbedaan jumlah pertumbuhan bakteri probiotik (Lactobacillus acidophilus dan Lactobacillus plantarum) dengan penambahan cookies prebiotik dan tanpa penambahan cookies prebiotik. Jumlah pertumbuhan Lactobacillus acidophilus dengan penambahan cookies prebiotik sebesar 13,92 log cfu/ml atau 8,5x1013 cfu/ml . Jumlah pertumbuhan Lactobacillus plantarum dengan penambahan cookies prebiotik sebesar 14,13 log cfu/ml atau 1,4x1014 cfu/ml, dimana jumlah tersebut sudah berada dalam kisaran jumlah bakteri Lactobacillus sp di dalam usus yaitu 1010-1012. Kesimpulan dari penelitian ini adalah cookies prebiotik efektif dalam meningkatkan pertumbuhan bakteri probiotik Lactobacilus acidophilus dan Lactobacillus plantarum

English Abstract

Children with Autism Spectrum Disorder (ASD) there is a condition called dysbiosis or an imbalance of intestinal microflora number which results a lower number of beneficial bacteria (probiotics) compared to a number harmful bacteria (pathogens). Prebiotics are foods that cannot be digested yet bring benefits by stimulating the growth of probiotic bacteria. Lactobacillus acidophilus and Lactobacillus plantarum are class of Lactic Acid Bacteria (BAL) which are commonly used as probiotic bacteria. Green beans and bananas are one of the foods that contain prebiotics and can be used to increase growth of probiotic bacteria such as Lactobacillus acidophilus and Lactobacillus plantarum. This research is an experimental design with two stages of preliminary and a stage of the main research. Preliminary stages are done to find out the content of starch resistant from prebiotic cookies meanwile the main research is done to test the growth of probiotic bacteria Lactobacillus acidophilus and Lactobacillus plantarum also to see the effectiveness of giving prebiotic cookies which is done by two treatments and two repetitions for each bacteria. Data were analyzed using Independent Sample T-test and showed a significantly differences between the amount of growth of probiotic bacteria (Lactobacillus acidophilus and Lactobacillus plantarum) with the addition of prebiotic cookies and without the addition of prebiotic cookies. The amount of growth of Lactobacillus acidophilus with the addition of prebiotic cookies is 13.92 log cfu / ml or 8,5x1013 cfu/ml . The amount of growth of Lactobacillus plantarum with the addition of prebiotic cookies is 14.13 log cfu / ml or 1,4x1014 cfu/ml, where the amount of it already in range of Lactobacillus sp bacteria in intestine is 1010-1012. Hence, this researched concluded that prebiotic cookies are effective to increase growth of probiotic bacteria Lactobacilus acidophilus and Lactobacillus plantarum
