Abstract
Endosulfan adalah insektisida golongan organoklorin yang memiliki efek toksik. Pada tahun 2011 Environmental Protection Agency (EPA) melaporkan endosulfan memiliki karakteristik yang persisten, bioakumulatif dan beracun. Penelitian di California menunjukan bahwa ibu hamil yang tinggal di daerah penggunaan endosulfan yang tinggi meningkatkan risiko yang tinggi melahirkan anak dengan autisme. Endosulfan juga dapat mengganggu neurotransmitter yaitu dopamin. Dopamin adalah neurotransmitter golongan monoamina yang berfungsi sebagai fungsi motorik, belajar, memori, pengolahan emosi dan kognisi sosial. Tujuan penelitian ini adalah menentukan pengaruh endosulfan selama kehamilan periode organogenesis terhadap kadar dopamin serum pada tikus putih strain wistar (Rattus norvegicus). Tikus betina yang sedang hamil dibagi menjadi 4 kelompok: kontrol (-), kontol (+) yang diberi endosulfan secara oral dengan dosis 0.07 ml, 0.33 ml, dan 1.6 ml. Pembedahan dilakukan pada hari ke- 30 setelah anak tikus dilahirkan, kemudian dilakukan pengukuran kadar dopamin serum menggunakan ELISA kit. Uji ANAVA menunjukkan paparan endosulfan selama kehamilan periode organogenesis terbukti secara signifikan menurunkan kadar dopamin serum tikus putih yaitu nilai p < 0.05. Disarankan perlu penelitian lebih lanjut untuk menentukan dosis maksimal dan membandingkan kadar dopamin serum induk tikus putih dengan anaknya.
English Abstract
Endosulfan is an insecticide from organochlorine group that has toxic effects. In 2011, the Environmental Protection Agency (EPA) reported that endosulfan has persistent, bioaccumulative and toxic characteristics. Research in California indicates that pregnant women living in areas with high use of endosulfan have higher risk of having a child with autism. Endosulfan can also disrupt neurotransmitters, such as dopamine. Dopamine is a monoamine neurotransmitter that functions as motor function, learning, memory, emotional processing and social cognition. The purpose of this study was to determine the effect of endosulfan during organogenesis period of pregnancy on serum levels of dopamine in Wistar strain rats (Rattus norvegicus). Pregnant female rats were divided into 4 groups : (-) control, (+) control that was given endosulfan orally at a dose of 0,07 ml, 0,33 ml, and 1.6 m. Dissection performed on the 30th day after the babies were born, then the serum levels of dopamine were measured using ELISA kits. ANOVA test showed that endosulfan exposure during organogenesis eriod of gestation significantly reduce serum levels of dopamine in the white rats with the value of p < 0.05. Further research is suggested to need to determine the maximum dose and to compare the serum levels of dopamine in the adult female white rats with their babies.
