Abstract

Tanah rhizosfer merupakan zona tempat berinteraksi mikroorganisme dengan akar tanaman, terdapat mikrooganisme yang menguntungkan untuk pertumbuhan tanaman. Mikroorganisme yang menguntungkan untuk meningkatkan pertumbuhan tanaman adalah kelompok rhizobakter atau Plant Growth Promoting Rhizobacteria (PGPR). Plant Growht Promoting Rhizobacteria memiliki peranan penting bagi tumbuhan dengan produksi fitohormon Indole Acetic Acid (IAA). Tujuan penelitian ini adalah mengetahui diversitas dan bakteri yang berpotensi sebagai penghasil IAA tanah rhizosfer Pinus (Pinus sp.) dari hutan Universitas Brawijaya serta mengidentifikasi isolat bakteri unggul berdasarkan sekuen 16S rDNA. Sampel tanah diambil dari tanah rhizosfer Pinus (Pinus sp.) Hutan Universitas Brawijaya. Bakteri tanah rhizosfer diisolasi dengan menggunakan media Tryptic Soy Agar yang ditambahkan triptofan 200 μg/mL dengan metode pour plate. Diversitas bakteri tanah rhizosfer dihitung dengan Indeks Diversitas Simpson. Bakteri penghasil IAA seleksi dengan uji kualitatif dan kuantitatif dengan Reagen Solkowski. Bakteri unggul penghasil IAA diidentifikasi berdasarkan sekuen 16S rDNA. Indeks diversitas bakteri tanah rhizosfer Pinus tergolong tinggi yaitu 0,92. Isolat UB 3.1 yang paling tinggi potensinya menghasilkan IAA sebesar 24,17 μg/mL dalam waktu inkubasi 72 jam. Berdasarkan sekuen 16S rDNA isolat UB 2.3 berkerabat dengan Myroides odoratimimus 6PE dengan nilai similaritas tertinggi 99,6 %.

English Abstract

Rhizosphere zone is site which microorganisms could interact with plant roots, there are some beneficial microoganisms for plant growth. One of microorganism that to increase plant growth is Plant Growth Promoting Rhizobacteria (PGPR). Plant Growth Promoting Rhizobacteria has an important role, such as Indole Acetic Acid (IAA). The purpose of this research is to know the diversity, the potency of IAA-producing bacteria and identifying bacterial isolates based on 16S rDNA sequences. The soil samples were taken from the Pine’s (Pinus sp.) rhizosphere at Brawijaya University Forest. The bacteria were isolated using Tryptic Soy Agar medium which contain trytophan 200 μg/mL pour plate method. The diversity of rhizosphere soil bacteria were calculated by the Simpson Diversity Index. The IAA producing bacteria were screened qualitative and quantitative assay with Salkowski Reagents. Indole Acetic Acid producing bacteria were identified based on 16S rDNA sequences. The diversity index of bacterial Pine’s rhizosphere soil was 0.92. The isolates of UB 3.1 has highest produced IAA 24.17 μg / mL. Based on the sequence of 16S rDNA UB 2.3 has relationship to Myroides odoratimimus 6PE with 99.6% similarity.
