Abstract

Fase premenopause dapat disebabkan oleh penurunan fungsi ovarium di dalam tubuh. Penurunan fungsi ovarium juga dapat disebabkan oleh meningkatnya kadar radikal bebas sehingga produksi estrogen dalam ovarium menjadi berkurang dan mempengaruhi proses folikulogenesis di dalam ovarium. Kemangi merupakan tanaman yang memiliki kandungan antioksidan dan fitoestrogen yang berfungsi dalam menangkal radikal bebas dan meningkatkan proses hormonal. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian serbuk daun kemangi (O. canum) terhadap folikulogenesis ovarium tikus putih (R. norvegicus) premenopause. Penelitian ini menggunakan 24 ekor tikus putih umur 11 bulan dan berat badan 180-220 g. Tikus di kelompokkan menjadi 4 perlakuan yaitu kontrol, pemberian larutan serbuk daun kemangi secara oral dengan dosis P1(0,25 g/kg BB); P2 (0,5 g/kg BB); P3 (1 g/kg BB). Pemberian larutan serbuk kemangi dilakukan setiap hari selama 15 hari yang dimulai pada fase estrus yang sama. Hari ke-16 tikus di bedah dan di ambil organ ovarium yang selanjutnya di buat preparat paraffin dengan pewarnaan Hematoxilin-Eosin. Pengamatan preparat menggunakan mikroskop cahaya BX51 dan hasil di analisis dengan analisis statistik ANOVA (Analysis of Variance). Hasil penelitian pemberian serbuk daun kemangi menunjukkan bahwa terjadi kecenderungan peningkatan jumlah folikel terhadap perlakuan kontrol yaitu pada dosis 3 folikel primordial, folikel tersier dan juga korpus luteum. Pemberian serbuk kemangi juga mempengaruhi perkembangan folikel primordial sampai folikel tersier karena kandungan flavonoid kemangi yang mampu meningkatkan perkembangan folikel yg mulai menurun akibat premenopause.

English Abstract

The premenopausal phase can be caused by decreased ovarian function in the body. Decreased ovarian function can also be caused by increased levels of free radicals so that estrogen production in the ovaries becomes reduced and affects the process of follicologenesis in the ovaries. Basil is a plant that contains antioxidants and phytoestrogens that work in warding off free radicals and increase hormonal processes. This study aims to determine the effect of basil leaf powder (O. canum) on premenopausal white ovarian folikulogenesis (R. norvegicus). This study used 24 white rat with age of 11 months and weight 180-220 g. Rats were grouped into 4 treatments, namely control, administration of oral basil dosage solution with a dose of P1 (0.25 g / kg BB); P2 (0.5 g / kg BB); P3 (1 g / kg BB). Provision of basil powder solution is done every day for 15 days starting at the same estrus phase. Day 16 of the rat in surgery and in the ovarian organ grab is subsequently made paraffin preparations with Hematoxilin-Eosin staining. The observations of the preparations used the BX51 light microscope and the results were analyzed by ANOVA statistical analysis (Analysis of Variance). The results of basil leaf powder on P1, P2 and P3 showed that there was a tendency of increasing the number of follicles on the control treatment at the dose of 3 primordial follicles, tertiary follicles and also the corpus luteum. The administration of basil powder also affects the development of primordial follicles to the tertiary follicles it is proved by a significant difference between primordial follicles and tertiary follicles based on statistical analysis.
