Abstract
Luka insisi adalah luka yang direncanakan dengan tepat dengan memperhatikan bentuk, arah, dan ukuran luka. Salah satu herbal penyembuh luka adalah kulit Apel Manalagi (Malus sylvetris mill) yang mengandung flavonoid dan Vitamin C. Penelitian ini bertujuan untuk mengetahui efek pemberian salep kulit apel manalagi (Malus sylvetris mill) terhadap pertumbuhan epidermis dan penurunan ekspresi TNF-α sebagai indikator penyembuhan luka . Salep kulit apel dibuat dengan vaselin album dan PGA (Pulvis Gummi Arabicum) dengan konsentrasi ekstrak kulit apel 25%, 35%, dan 45%. Hewan coba dalam penelitian ini adalah tikus (Rattus norvegicus) jantan dengan berat 150-250 gram berumur 8-12 minggu yang dibagi menjadi 5 kelompok. Kelompok 1 adalah kelompok kontrol negatif yang tidak diberi perlakuan. Kelompok 2 adalah kelompok kontrol positif yang diberi perlakuan insisi namun tidak diberi terapi. Kelompok 3, 4 dan 5 adalah kelompok yang dinsisi dan diterapi dengan salep kulit apel dengan dosis bertingkat 25%, 35%, dan 45%. Parameter yang diukur dalam penelitian ini adalah pertumbuhan epidermis yang diamati secara histopatologi dan ekspresi TNF-α yang diukur menggunakan Imunohistokimia. Analisis data dilakukan dengan cara deskriptif dan uji One Way ANOVA dan uji lanjutan Tukey dengan tingkat kepercayaan 95% (α= 0,05). Pemberian salep kulit apel manalagi konsentrasi 25%, 35%, dan 45% menunjukkan perbaikan pada pertumbuhan pada epidermis. Terapi salep kulit apel manalagi (Malus sylvetris mill) konsentrasi 45% menunjukkan penurunan yang signifikan terhadap ekspresi TNF-α di jaringan kulit tikus (Rattus norvegicus) pasca insisi.
English Abstract
Incision wound is wound with planned in shape, direction, and size of the wound. This study aims to determine the effect of the Manalagi Apples (Malus sylvestris mill) dregs ointment to support growth of epidermis and decreased expression of TNF-α as the healing of the incision. The apple ointment was made of vaselin album and PGA base with three types of apple extract concentration 25%, 35%, and 45%. The animal model in this research were 150-250 g, eight until twelve week-old male rat (Rattus norvegicus). The rat were divided into five treatment groups. Each group contains four rats. The first group was the negative control which hadn’t anything treatment. The second group was the positive control group, which was had insicion without therapy. The third, fourth, fifth groups were therapy group, which was had incision and underwent therapy with 25%, 35%, and 45% apple ointment. The parameters in this research are growth of epidermis observed with histopathology method and expresion of TNF-α measured by Imunohistokimia. The data was analyzed with One way ANOVA test and Tukey range test with 95% confidence level (α=0,05). All of the therapy group 25%, 35%, and 45% apple ointment showed increase the growth of epidermis. The fifth group (45% apple ointment) showed decrease of TNF-α expression in skin tissue of rat.
