Abstract
Asap rokok dapat menurunkan level antioksidan, meningkatkan pelepasan radikal superoksida, dan menyebabkan nekrosis akibat stimulasi dari TNF-α. Jantung merupakan organ penting dalam tubuh karena fungsinya sebagai sistem kardiovaskuler. Ekstrak etanol propolis lebah Trigona sp diketahui mengandung Caffeic Acid Phenethyl Ester (CAPE) yang bekerja sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui histopatologi jantung dan ekspresi TNF-α pada tikus (Rattus norvegicus) hasil paparan asap rokok yang diberi ekstrak propolis lebah Trigona sp sebagai preventif. Hewan coba yang digunakan adalah tikus putih strain Wistar. Tikus dibagi menjadi lima kelompok yaitu kelompok kontrol negatif (K-), kelompok kontrol positif (K+), kelompok tikus P1, P2, P3 yang masing-masing diberi ekstrak etanol propolis lebah Trigona sp 10mg/200g/hari, 20mg/200g/hari, 30mg/200g/hari kemudian diberi paparan asap rokok 2 batang/hari selama 14 hari. Pengukuran ekspresi TNF-α menggunakan uji Imunohistokimia, analisa data dilakukan secara kuantitatif menggunakan uji One Way ANOVA dan dilanjutkan dengan uji Tukey (P<0,05). Histopatologi jantung menggunakan pewarnaan Hematoxylin Eosin (HE). Hasil gambaran histopatologi organ jantung dianalisa secara deskriptif. Hasil penelitian menunjukkan ekstrak propolis lebah Trigona sp secara signifikan (P<0,05) mencegah peningkatan ekspresi TNF-α pada tikus yang terpapar asap rokok, dengan dosis terbaik untuk mencegah peningkatan ekspresi TNF-α adalah 30mg/200g/hari. Terapi ekstrak etanol propolis lebah Trigona sp mencegah kerusakan kondisi histopatologi sel otot jantung. Kesimpulan dari penelitian, ekstrak etanol propolis lebah Trigona sp mampu mencegah peningkatan ekspresi TNF-α dan dapat mencegah kerusakan otot jantung pada tikus yang dipapar asap rokok.
English Abstract
Cigarette smoke could reduce antioxidant levels, increase the release of superoxide radical, and emerge the necrosis because of the stimulation of TNF- α. Heart is premier organ in the body, it is included into the cardiovascular system.The ethanol extract of propolis bee Trigona sp is known to contain Caffeic Acid Phenethyl Ester (CAPE) which acts as an antioxidant. This research was aims to determine the histopathology of heart and expression of TNF-α in rat (Rattus norvegicus) exposure to cigarette smoke extracted with Trigona sp extract as preventive. The experimental animal used was Wistar strain white rat. The study consisted to five groups in which contained of negative control (K-), positive control (K+), groups of rats P1, P2, P3 were given ethanol extract of propolis bee Trigona sp 10mg/200g/day, 20mg/200g/day, 30mg/200g/day and those were exposed to cigarettes smoke as much 2 cigarettes/day within 14 days. The measurement of expression TNF-α used Immunohistochemistry tests, quantitative data analysis performed was used One Way ANOVA followed by Tukey's test (P <0.05). Histopathology of heart used Hematoxylin Eosin staining (HE). The results of overview heart organ histopathology were analysed descriptively. The results showed that Trigona sp extract significantly (P<0.05) prevented the increase of TNF-α expression in rat exposed to cigarette smoke, the best dose to prevent the increase of TNF-α expression was 30mg/200g/day. The therapy using ethanol extract of propolis bee Trigona sp prevents the damage histopathological condition of heart muscle cells. The conclusions from the research, ethanol extract of propolis bee Trigona sp is able to prevent the increase of TNF-α expression and can prevent the damage of heart muscle in rat exposed to cigarette smoke.
