Abstract

Traumatic Brain Injury (TBI) adalah cedera pada otak akibat benturan, pukulan, dan gonjangan pada kepala. Ekstrak pegagan mengandung Asiatikosida yang mampu menginisiasi neurogenesis melalui aktivasi siklus kreb yang ditandai dengan SOX2. Hidrogel fitosom ekstrak pegagan merupakan drug delivery systems yang memperbaik farmakokinetik dan farmakodinamik suatu zat. Penelitian ini bertujuan untuk mengetahui efek pemberian hidrogel fitosom ekstrak pegagan terhadap peningkatan SOX2 pada tikus model TBI. Desain penelitian ini menggunakan true experiment design dengan rancangan randomized post test only controlled group design yang menggunakan delapan kelompok tikus Sprague dawley jantan yang masing-masing terdiri dari tiga sampel. Variabel yang diukur adalah SOX2 yang dilihat secara mikroskopis dengan pengecatan imunohistokimia. Hasil penelitian menunjukkan bahwa terdapat perbedaan yang signifikan antara kelompok kontrol hari ke-1 terhadap kelompok hidrogel fitosom hari ke-1 (p= 0,043), antara kelompok kontrol hari ke-3 terhadap kelompok hidrogel fitosom hari ke-3 (p= 0,046), antara kelompok kontrol hari ke-7 terhadap kelompok hidrogel fitosom hari ke-7 (p= 0,046), dan antara kelompok kontrol hari ke-14 terhadap kelompok hidrogel fitosom hari ke-14 (p= 0,046). Dari hasil analisis data, dapat disimpulkan bahwa pemberian hidrogel fitosom ekstrak pegagan mampu memodulasi neurogenesis yang ditandai peningkatkan ekspresi SOX2 pada tikus model traumatic brain injury

English Abstract

Traumatic Brain Injury (TBI) is an injury to the brain due to impact, blow, and shock on the head. Pegagan extract contains Asiaticoside which is able to initiate neurogenesis through activation of creb cycle, characterized by SOX2. Hydrogel fitosom Pegagan extract is a drug delivery system that improves pharmacokinetics and pharmacodynamics of a substance. This study aims to determine the effect of hydrogel fitosom pegagan extract on the increasing of SOX2 in mouse model of TBI. The design of this study use true experiment design with randomized post test only controlled group design using eight groups of male Sprague dawley mouse, each consisting of three samples. The measured variable is SOX2 which is seen microscopically by imunohistochemical. The results showed that there was a significant difference between control group on 1st day and hydrogel fitosome pegagan ekstrak group on 1st day (p = 0,043), between control group on 3rd day and hydrogel fitosome pegagan ekstrak group on 3rd day (p = 0,046), control group on 7th day and hydrogel fitosome pegagan ekstrak group on 7th day (p = 0.046), and between control group on 14th day and hydrogel fitosome pegagan ekstrak group on 14th day (p = 0.046). From the results of data analysis, it can be concluded that the administration of hydrogel phytosome pegagan extract is able to modulate neurogenesis in traumatic brain injury model mouse, marked by enhanced expression of SOX2.
