Abstract
Fibrosis merupakan penyakit progresif yang diakibatkan dari proses perbaikan luka abnormal. Proses perbaikan luka melibatkan berbagai peran sitokin terutama TGF-β. Bleomycin merupakan agen kemoterapi antibiotik anti tumor dan agen standard untuk induksi hewan model fibrosis paru. Kefir adalah susu fermentasi mengandung bakteri dan jamur yang memiliki berbagai keuntungan kesehatan termasuk sebagai imunomodulator. Studi ini bertujuan untuk mengetahui peran kefir sebagai imunomodulator dalam menurunkan TGF- β pada tikus model fibrosis subkronis. Studi ini merupakan eksperimental menggunakan tikus Wistar jantan yang telah diinduksi bleomycin 9 kali secara intranasal terbagi dalam empat kelompok yaitu kelompok kontrol (tanpa pemberian kefir), P1 (kefir 2.5 ml/grBB/hari), P2 (3.5 ml/grBB/hari) dan P3 (4.5 ml/grBB/hari) selama 30 hari. Kadar TGF-β serum diukur dengan metode ELISA. Hasil penelitian memperlihatkan bahwa kadar TGF-β terendah yaitu kelompok tikus yang dipapar kefir 4.5 ml/grBB/hari (P3). Sedangkan, kadar TGF-β tertinggi yaitu kelompok tikus yang dipapar kefir 2.5 ml/grBB/hari (P1). Namun, perbedaan kadar TGF-β antar kelompok tidak signifikan (p>0.05). Dengan demikian, uji peran immunomodulator kefir tidak terbukti signifikan pada ekspresi TGF-β pada model tikus fibrosis subkronis.
English Abstract
Fibrosis is a progressive disease resulting from an abnormal wound healing process. The wound healing process involves the various roles of cytokines, especially TGF-β. Bleomycin is a chemotherapeutic antitumor antibiotic agent and the standard agent for induction of pulmonary fibrosis in animal models. Kefir is a fermented milk that contains bacteria and fungi, which have various health benefits, including as an immunomodulator. This study aims to evaluate the role of kefir as an immunomodulator in down-regulated TGF-β in a mouse model of subchronic fibrosis. This study was experimental using male mouse Wistar, which has induced by bleomycin 9 times via intranasal are divided among four groups such as, control group (without kefir), P1 (kefir 2.5 ml/grBB/day), P2 (3.5 ml/grBB/day) and P3 (4.5 ml/grBB/day) for 30 days. Serum TGF-β levels were measured by the ELISA method. The results showed that the lowest level of TGF-β was the mouse group with exposed to kefir 4.5 ml/grBB/day (P3). Meanwhile, the highest level of TGF-β was mouse group exposed to kefir 2.5 ml/grBB/day (P1). However, the difference level TGF-β between groups was not significant (p>0.05). Thus, the role test of immunomodulatory kefir was not proven significantly in TGF-β expression in mouse model of subchronic fibrosis.
