Abstract

Jantung merupakan organ yang berperan penting dalam tubuh untuk memompa darah. Gangguan terhadap jantung dapat menyebabkan kecacatan hingga kematian. Contoh penyebab gangguan jantung ialah Reactive Oxygen Species (ROS). Gangguan akibat ROS dapat dicegah dengan antioksidan. Salah satu sumber antioksidan ialah kelor (Moringa oleifera). Tumbuhan ini mudah dijumpai dan telah banyak dimanfaatkan baik sebagai makanan maupun obat. Dalam penelitian ini model yang digunakan ialah zebrafish, karena perkembangannya cepat, mudah diamati, memiliki genetik serta metabolisme yang mirip dengan manusia dan sensitif terhadap pemberian zat kimia. Penelitian ini menggunakan post-test only control group design yang bertujuan untuk mengetahui pengaruh ekstrak daun kelor terhadap heart rate embrio zebrafish. Sampel berupa telur zebrafish berusia 2 hpf (hour post fertilization) hasil perkawinan induk dewasa, yang dipilih dengan cara simple random sampling dan dibagi menjadi 4 kelompok, yaitu kelompok kontrol, kelompok ekstrak daun kelor 0,56 ppm (part per million), 1,12 ppm dan 2,24 ppm. Telur dipapar dengan ekstrak daun kelor hingga berusia 72 hpf. Variabel yang diamati ialah heart rate embrio zebrafish yang berusia 72 hpf. Hasil penelitian menunjukan bahwa pemberian ekstrak daun kelor mampu meningkatkan heart rate embrio zebrafish secara signifikan dibandingkan dengan kelompok kontrol (p>0.05). Namun tidak ada perbedaan heart rate yang signifikan dengan peningkatan dosis yang diberikan. Kesimpulan dari penelitian ini ialah pemberian ekstrak daun kelor meningkatkan heart rate embrio zebrafish.

English Abstract

Heart is an important organ of the body to pump the blood. Any disturbance to the heart can cause morbidity or mortality. The examples of heart diorders causes is Reactive Oxygen Species (ROS). The bad effects caused by ROS can be prevented by antioxidants. One source of antioxidants is moringa (Moringa oleifera). This plant is easy to find and has been widely used both as food and medicine. In this study, the model used was zebrafish, because its development is fast, can be observed easily, has a genetic and metabolism similar to humans and sensitive to the giving of chemicals. This study used laboratory experimental with post-test only control group design which purpose to determine the effect of moringa leaf extract on zebrafish embryonic heart rate. The samples was zebrafish eggs from adult mating, selected by simple random sampling and divided into 4 groups. One group as a control group, and 3 treatment groups that exposed to moringa leaf extract with a doses of 0,56 ppm (part per million), 1,12 ppm and 2,24 ppm. Eggs exposed with Moringa leaf extract up to 72 hpf (hour post fertilization). The observed variable was a 72 hpf zebrafish embryonic heart rate. The result of this study showed that giving moringa leaf extract was able to increase the heart rate of zebrafish embryo significantly compared with the control group (p> 0.05). However, there was no significant difference in heart rate with increased doses. The conclusion of this study is the extract of moringa leaf increase the heart rate of zebrafish embryo
