Abstract

Kulit pisang kepok kuning berpotensi menjadi bahan aditif dalam produk kosmetik, karena mengandung senyawa fitokimia seperti tannin, flavonoid, saponin, dan alkaloid yang memiliki daya antibakteri terhadap Staphylococcus aureus. Pada proses ekstraksi soxhlet kulit pisang yang bertujuan untuk mengambil senyawa fitokimia, lamanya waktu penundaan proses pengeringan kulit pisang kapok kuning dan lamanya waktu ekstraksi dapat mempengaruhi kandungan senyawa antibakteri di dalamnya. Penelitian ini bertujuan untuk mengetahui pengaruh waktu penundaan dan siklus ekstraksi kulit pisang kepok kuning terhadap kandungan senyawa antibakteri dan daya antibakteri Staphylococcus aureus. Kulit pisang diberi perlakuan penundaan proses pengeringan selama 12, 24, 48,dan 72 jam. Proses sokhletasi kulit pisang dilakukan selama 4 ,6, dan 8 siklus dengan menggunakan pelarut ethanol, kemudian dipisahkan menggunakan rotary evaporator. Kandungan fitokimia dalam ekstrak diidentifikasi menggunakan uji fitokimia dan FT-IR. Sedangkan, daya antibakteri ekstrak kulit pisang terhadap Staphylococcus aureus diuji menggunakan metode disc diffussion. Hasil penelitian menunjukkan bahwa penambahan waktu tunda dan jumlah siklus tidak berpengaruh secara signifikan terhadap jenis komponen senyawa antibakteri di dalam ekstrak kulit pisang kepok kuning, dimana senyawa antibakteri yang dapat terekstrak adalah tanin, flavonoid, dan saponin. Selain itu, daya antibakteri yang paling kuat terhadap Staphylococcus aureus adalah ekstrak kulit pisang kepok kuning pada variabel 12 jam dan 4 siklus. Sedangkan, daya antibakteri yang paling lemah adalah 72 jam dan 8 siklus. Sehingga, semakin lama waktu tunda dan semakin besar siklus ekstraksinya maka daya antibakteri ekstrak kulit pisang kepok kuning akan mengalami penurunan.

English Abstract

In Indonesia, yellow kepok banana peel only used as animal feed. Whereas banana peels contain phytochemical compounds such as tannins, flavonoids, saponins and alkaloids which have antibacterial activity against Staphylococcus aureus, so banana peels have the potential to be additive in cosmetic products. In the process of soxhlet extraction of banana peel which aims to take phytochemical compounds, the length of time delay of yellow banana peel drying process and the length of extraction time may affect the content of antibacterial compounds in it. This study is aimed to understand the effect of delay time and extraction cycles of yellow kepok banana peel against antibacterial compounds and antibacterial activity of Staphylococcus aureus. Banana peel was treated with a delay of drying process for 12, 24, 48, and 72 hours. Banana peel soxhletation was carried out for 4, 6 and 8 cycles using ethanol as solvent. Banana peel-ethanol extract was then separated using rotary evaporator. Phytochemical content in the extract was identified using phytochemical test and FT-IR. Whereas, antibacterial activity of banana peel extract against Staphylococcus aureus was tested using disc diffussion method. The results showed that the increase of extraction cycle and the longer delay time caused differences in the content of the antibacterial compound. Increasing the number of cycles will increase the solubility of tannins, whereas the solubility of flavonoids and saponins will be reduced because of the decomposition reaction at 80 °C. Increasing the delay time will reduce the content of tannins and flavonoids due to enzymatic browning reaction, while saponin compound will increase due to softening reaction. The results showed the increase of extraction cycle and the longer delay time does not significantly affect the antibacterial compound content in the yellow kepok banana peel extract, wherein the antibacterial compound that can be extracted is tannins, flavonoids and saponins. Moreover, the most powerful antibacterial activity against Staphylococcus aureus is yellow kepok banana peel extract with variable delay time 12 hours and 4 cycles. Meanwhile, the weakest antibacterial activity is yellow kepok banana peel extract with variable delay time 72 hours and 8 cycles. Thus, the longer the delay time and the greater the cycle of extraction then the antibacterial activity of yellow kepok banana peel extract will decrease.
