Abstract

Sepsis adalah manifestasi sindroma klinis sebagai akibat proses inflamasi dan respon tubuh berlebihan terhadap infeksi mikroorganisme atau toksinnya. Penyebab sepsis paling banyak adalah bakteri gram negatif yaitu sekitar 60-70% dari keseluruhan kasus. Escherichia coli merupakan salah satu bakteri gram negatif penyebab sepsis yang menghasilkan endotoksin dari bagian membran sel nya yaitu berupa lipopolisakarida (LPS). Tujuan dari penelitian ini adalah untuk mengetahui efek pemberian ekstrak ragi (Saccharomyces cerevisiae) untuk menurunkan kondisi sepsis, yang diamati melalui pengukuran kadar superoksida dismutase (SOD) dengan metode spektrofotometri dan gambaran histopatologi hepar pada tikus model sepsis hasil induksi Escherichia coli. Hasil pengukuran SOD dianalisa menggunakan OneWay ANOVA (α=0,05) dan analisa lanjutan menggunakan uji BNT, sedangkan gambaran histopatologi hepar dengan analisa deskriptif. Penelitian ini menggunakan rancangan acak lengkap (RAL) dengan yang terdiri dari lima kelompok, yaitu kontrol negatif, kontrol positif (diinfeksi Escherichia coli) dan tiga kelompok terapi (diinfeksi Escherichia coli dan diterapi dengan ekstrak ragi dosis bertingkat yaitu 10 mg/kgBB, 20 mg/kgBB dan 30 mg/kgBB). Induksi Escherichia coli dengan dosis 1,5 x cfu/ml diinjeksikan secara intraperitoneal sebanyak 1 ml pada hari kedelapan penelitian. Terapi ekstrak ragi diberikan secara per-oral enam jam pasca induksi, dan dilanjutkan setiap 24 jam selama lima hari. Hasil dari penelitian ini menunjukkan bahwa pemberian terapi ekstrak ragi dapat meningkatkan kadar SOD dan perbaikan gambaran histopatologi hepar pada hewan coba model sepsis

English Abstract

Sepsis is a manifestation of clinical syndrome as a result of inflammatory processes and excessive body response to microorganisms infection or toxins. The most common cause of sepsis is gram-negative bacteria that is about 60-70% of all cases. Escherichia coli is one of the gram-negative bacteria causing sepsis that produce endotoxin calls lipopolysaccharide (LPS). The purpose of this study is to investigate the effect of yeast extract (Saccharomyces cerevisiae) to reduce sepsis condition, which was observed by measurement of superoxide dismutase (SOD) by spectrophotometric method and histopathologic image of liver in mouse model of induction of Escherichia coli induction. The results of SOD measurements were analyzed using OneWay ANOVA (α = 0.05) and continued analysis using BNT test, while histopathologic description of liver with descriptive analysis. This study used a completely randomized design (CRD) with five groups: negative control, positive control (infected Escherichia coli) and three treatment groups (infected Escherichia coli and treated with 10 mg / kg BW, 20 mg / kg BW and 30 mg / kg BW dose yeast extract). The induction of Escherichia coli with a dose of 1,5 x cfu/ml was injected intraperitoneally 1 ml on the eighth day of the study. Yeast extract therapy is administered on a per-oral basis six hours post induction, and continued every 24 hours for five days. The results of this study showed that the treatment of yeast extract could increase SOD level and improve histopathologic image of liver in animal model sepsis.
