Abstract
Sedikit tahu daun Padina australis sebagai kering dan teh. Tujuan penelitian adalah mengetahui karakteristik daun alga coklat Padina australis dalam bentuk kering dan teh yang dibandingkan dengan standar kualitas teh berdasarkan SNI nomor (3836:2013). Padina australis yaitu diangin-anginkan (suhu 37ºC, 48 jam, RH 70%) juga dalam bentuk teh, direndam Ca(OH)2, pH 11, selama 6 jam dan dikeringkan oven suhu 80oC, 20 menit). Produk ini kemudian dikarakterisasi senyawa bioaktif teh (kuersetin) menggunakan LC-MS, FTIR, aktivitas antioksidan, kandungan protein sebagai bahan makanan. Sebagai produk teh, hasil penelitian menunjukkan bahwa kadar polifenol (10.89 mgGAE/100gr), logam berat (kadmium (0,033 ppm), timbal (0,002 ppm), timah (0,093 ppm), merkuri dan arsen (tidak terdeteksi), dan kapang (1 x 102) memenuhi Standar Nasional Indonesia namun tidak pada kandungan air (10.459%), abu (45.272%), serat kasar (18.41%), dan standar mikrobiologi (coliform 1,1x105 AMP/g dan TPC 18x103 koloni/g). Antioksidan IC50 sebesar 427.69 ppm, hasil FTIR teh diduga terdapat ikatan gugus hidroksil (OH), CH alifatik, C=C aromatik dan C-O mengindikasikan senyawa golongan flavonoid, dari hasil LC-MS terdeteksi kuersetin dalam kandungan yang cukup kecil. Sebagai bahan pangan memiliki protein 6.03%. Sebagai produk kering, penelitian tersebut menunjukkan bahwa loga berat (kadmium (0,035 ppm), timbal (0,001 ppm), timah (0,086 ppm), merkuri dan arsen (tidak terdeteksi), dan kapang (1 x 102) memenuhi Standar Nasional Indonesia namun tidak pada kandungan air (13.107%), abu (14.377%), serat kasar (18.32%), dan standar mikrobiologi (coliform 1,1x105 AMP/g dan TPC 29x103 koloni/g). Antioksidan IC50 sebesar 770.77 ppm, hasil FTIR kering menunjukkan bahwa mengandung komponen sulfur dan halogen, dari hasil LC-MS terdeteksi kuersetin dalam kandngan yang cukup kecil. Sebagai bahan pangan memiliki protein 7.10%.
English Abstract
A little know of Padina australis leaves as dried and dried tea. The purpose of this research was know the characteristic of dry and tea brown algae leaves of Padina australis compared to tea quality standard based on Indonesian National Product Standards number (3836: 2013). Padina australis was dried in dry air (temperature 37ºC, 48 hours, RH 70%) as well on tea, except soaked in Ca(OH)2, pH 11, 6 hours then oven dried 80ºC, 20 minutes. These products then to be characteristation in tea quality bioactive compound (quercetin) using LC-MS, FTIR, antioxidant activity, protein content as food ingredient. As tea product the research showed that polyphenol contennt (10.89mgGAE/100gr), heavy metals (cadmium (0.033 ppm), lead (0.093 ppm), tin (0.002 ppm), mercury and arsenic (undetectable)) value, mold content (1x102) were fulfill with Indonesian National Product Standards but not in water (10.459%, ash (45.272%, fiber (18.41%) content, and microbiology standard (coliform (1.1x105 APM/g) and TPC (18x103 koloni/g)). IC50 of antioxidant were 427.69 ppm, from FTIR it showed group hydroxyl (OH), CH aliphatic, C=C aromatic and C-O that indicated group of flavonoid compound, from LC-MS it showed quercetin in small enough content. As food ingredient, it had 6.03% of proteins.
