Abstract

Cabai paprika (Capsicum annum .L. var. grossum) merupakan tanaman sayuran yang mulai dikenal di Indonesia pada tahun 1990-an. Di Eropa dan Amerika paprika banyak digunakan sebagai penyedap makanan. Penanaman paprika semakin berkembang akibat meningkatnya kebutuhan pasar sehingga memiliki prospek yang cerah untuk di budidayakan. PMMoV (Pepper Mild Mottle Virus) merupakan virus yang dikategorikan sebagai seedborne pada C. annuum dan C. frutescens, Masalah kesehatan benih merupakan hal yang penting untuk dijaga agar tidak mempengaruhi produksi tanaman paprika. Oleh karena itu diperlukan pengujian kesehatan benih dan mengkaji cara yang efektif untuk mengeleminasi virus yang terbawa benih, mengingat kebutuhan untuk persyaratan distribusi benih harus terbebas dari spesifik patogen target. Tujuan dari penelitian ini adalah untuk mengkaji perlakuan yang efektif untuk pengendalian penyakit pada benih Paprika (C. annum. L. var.grossum). Hipotesa pada penelitian ini ialah PMMoV yang menginfeksi benih Paprika (C. annum. L. var.grossum) dapat tereleminasi dengan perlakuan fisik, kimia atau senyawa nabati. Penelitian dilakukan di Laboratorium Proteksi Tanaman, Laboratorium Molekuler Departemen Bioteknologi PT. BISI Internasional, Tbk dan green house di Dusun Jungo, Desa Bumiaji, Kota Batu di mulai dari Desember 2016 sampai dengan Mei 2017. Metode pengujian efektivitas perlakuan pada penelitian ini menggunakan Double Antibody Sandwich Enzyme-linked immunosorbent assay (DAS- ELISA) dan Reverse Transciptase Polymerase Chain Reaction (RT- PCR). Penelitian terdiri dari 6 perlakuan, dengan masing- masing perlakuan diulang sebanyak 4 ulangan. Parameter yang diamati ialah hasil DAS- ELISA, Uji Grow Out Test (GOT) meliputi daya kecambah dan vigor dan Uji gejala tanaman meliputi masa inkubasi dan kejadian penyakit. Data pengamatan dianalisis mengunakan analisis ragam pada taraf 5 %. Bila hasil pengujian terdapat perbedaan yang nyata maka dilanjutkan dengan uji BNT pada taraf 5 %. Hasil pengujian Double Antibody Sandwich Enzyme-linked immunosorbent assay (DAS- ELISA) menunjukkan perlakuan perendaman benih paprika dalam ekstrak minyak daun cengkeh 4% selama 20 menit dan Trisodium phosphate (TSP) 9.6% selama 2 jam efektif menghilangkan virus pada benih cabai paprika. Berdasarkan hasil analisis ragam menunjukkan bahwa semua perlakuan dapat menurunkan daya perkecambahan dibandingkan dengan kontrol, Perlakuan perendaman benih dengan TSP 9.6% selama 2 jam direkomendasikan karena memiliki daya kecambah sebesar 88.3%. Berdasarkan hasil analisis ragam semua perlakuan tidak berbeda nyata terhadap kejadian penyakit namun perlakuan perendaman benih dengan TSP 9.6% selama 2 jam memiliki presentase kejadian penyakit terkecil (30.0%) dibandingkan dengan perlakuan lainya.

English Abstract

Bell Pepper (Capsicum annuum L. var. grossum) is a horticultural product and it started to be popular in Indonesia in 1990s. In Europe and America, bell peppers are utilized as food spices. Cultivation of bell peppers come to an increase as the result of their rising demand, and thus it becomes a prospect of profit. PMMoV (Pepper Mild Mottle Virus) is categorized as seed-borne plant virus on C. annum and C. frutescens. Seed health issue is an important thing. Thus, it is also important to assess seed health and figure out and effective way of seed-borne plant virus elimination, as it is required to distribute virus-free seeds. The objective of this reseach was to figure out an effective treatment for bell peppers’ disease management. The hypothesis tested in this research was PMMoV infecting bell pepper seed can be eliminated with physical, chemical, or natural compund treatment. This research was conducted in Plant Protection Laboratory, Molecular Laboratory of Biotechnology Department of PT BISI Internasional, Tbk., and a greenhouse in Jungo Hamlet, Bumiaji Village, Batu City from December 2016 up to May 2017. The effectiveness test in this research consisted of Double Antibody Sandwich Enzyme-linked Immune Sorbent Assay (DAS- ELISA) and Reverse Transciptase Polymerase Chain Reaction (RT- PCR). There were 6 treatments in this experiment, with 4 repetitions for each one. Observation parameters were DAS-ELISA results, Grow Out Test (GOT) consisting seed viability and seed vigor index, also, plant symptom test consisting disease incubation period and incidence. Observation data were analyzed with variance analysis. The data were further analyzed with 5% level of Least Significant Difference test if they appeared to be significantly different. Results of DAS-ELISA test showed that treatment immersion in 4% of clover leave extract for 20 minutes and immersion in 9.6 % of trisodium phosphate for 2 hours were effective to eliminate seed-borne virus. According to variance analysis results, all treatments decreased seed viability, compared to control. Treatment P4 could be recommended as it showed 88,3% of seed viability. Variance analysis results showed that all treatments were not significantly different in terms of plant disease incidence. However, treatment P4 proved to have the lowest disease incidence (30.0 %) compared to others.
