Abstract
Bulu Babi merupakan salah satu komoditas perikanan yang cukup populer di masyarakat. Banyak penelitian tentang bulu babi sebagai antibakteri,antitumor,antikanker. Didalamnya sangat bermanfaat dibidang pengobatan sebagai antibiotic tipe baru . Penelitian ini bertujuan untuk mendapatkan ekstrak bulu babi menggunakan metode maserasi, menentukan aktivitas antibakteri menggunakan metode difusi cakram ; dan menentukan berat moleku protein ekstrak bulu babi Diadema setosum menggunakan Elektroforisis SDS-PAGE. Bulu babi diperoleh dari pantai pasir Putih Situbondo sebanyak 5 kilogram. Ekstrak gonad bulu babi memiliki aktivitas antibakteri tertinggi dengan zona hambat (0,96 ± 0,03) mm terhadap bakteri Salmonella typhi dan (0,86 ± 0,02) mm terhadap bakteri Streptococcus pyogenes. Hasil penelitian diolah menggunakan Rancangan Acak Lengkap Sederhana dengan 4 kali ulangan dan jika didapatkan hasil yang berbeda nyata maka dilakukan uji BNT. Hasil penelitian menunjukkan hasil terbaik pada penambahan konsentrasi ekstrak 8000 ppm. Didapatkan berat molekul dalam uji Elektroforisis SDS-PAGE sebesar 17,6 kDa -260,0 kDa
English Abstract
Sea Urchin is one of the fishery commodities that are quite popular in the community. Many studies of sea urchins as antibacterial, antitumor, anticancer. In it is very useful in the field of medicine as a new type of antibiotic. The aim of this study was to obtain the extract of sea urchins using maseration method, to determine the antibacterial activity using disc diffusion method; And determine the molecular weight of the protein extract of the Diadema setosum using Elektroforisis SDS-PAGE. Pig hair is obtained from White sand beach Situbondo as much as 5 pounds. Gonad extract has the highest antibacterial activity with inhibit zone (0.96 ± 0.03) mm against Salmonella typhi bacteria and (0.86 ± 0.02) mm against Streptococcus pyogenes bacteria. The results were processed using Simple Completely Randomized Design with 4 replications and if the results obtained were significantly different then BNT test was performed. The results showed the best results on the addition of extract concentration of 8000 ppm. The molecular weight obtained in the Electrophoresis SDS-PAGE test was 17.6 kDa -260.0 kDa
