Abstract

Penelitian ini dirancang untuk mengetahui perbedaan kualitas level BSA 0,2% dan 0,4% dalam pengencer CEP-2 + 10% Kuning telur. Semen segar dikoleksi dari 3 pejantan Sapi PO berumur 3 tahun menggunakan vagina buatan dilakukan satu kali seminggu. Metode penelitian ini adalah experimental laboratorium menggunakan pola Rancangan Acak Kelompok (RAK) dengan tiga perlakuan dan sepuluh ulangan, perlakuan adalah menggunakan pengencer CEP-2 + 10% Kuning Telur (P0), CEP-2 + 10% Kuning Telur + 0,2% BSA (P1), CEP-2 + 10% Kuning Telur + 0,4% BSA (P2). Apabila terdapat perbedaan yang nyata maupun sangat nyata akan dilanjutkan dengan Uji Jarak Berganda Duncan (UJBD). Hasilnya menunjukkan bahwa pengencer CEP-2 – 10% kuning telur dengan 0,2% BSA berbeda nyata (p<0,05) terhadap kualitas spermatozoa pada PTM selama proses pembekuan P0 (75,35±4,51)%, P1 (82,08±5,41)% dan P2 (79,73±4,79)% dalam hal persentase viabilitas spermatozoa. Semua perlakuan total spermatozoa motil pada PTM selama proses pembekuan lebih tinggi dibandingkan standar SNI 40 juta/ml. Kesimpulan dari penelitian ini adalah 0,2% BSA efektif menjaga persentase viabilitas selama proses pembekuan, peneliti menyarankan untuk menggunakan 0,2% BSA karena mampu mempertahankan kualitas spermatozoa selama proses pembekuan.

English Abstract

This study was designed to determine the difference quality of BSA level 0.2% and 0.4% in CEP-2 + 10% egg yolk. Fresh semen was collected from 3 filial ongole bull aged 3 years using artificial vagina once a week. The method of this research experimental laboratory using Randomized Block Design (RBD) with three treatments and ten replications. The treatment was using CEP-2 + 10% egg yolk (P0), CEP-2 + 10% egg yolk + 0.2% BSA (P1), CEP-2 + 10% egg yolk + 0.4% BSA (P2). If there was a real or very real difference, it will be continued with Duncan Multiple Range Test. The results showed that the CEP-2 + 10% yolk with 0.2% BSA differs significantly (P<0.05) on spermatozoa quality at PTM during freezing P0 (75.35 ± 4.51)%, P1 (82 , 08 ± 5.41)% and P2 (79.73 ± 4.79)% in terms of percentage viability of spermatozoa. All total motile spermatozoa treatment at PTM during freezing process was higher than SNI standard 40 million/ml. The conclusion was that 0.2% of BSA was effective in maintaining the percentage of viability during the freezing process. The suggestion used is 0.2% BSA because it was able to maintain the quality of spermatozoa during the freezing process.
