Abstract

Diabetes Melitus (DM) tipe I merupakan gangguan sistem endokrin yang ditandai penurunan produksi insulin akibat kerusakan sel β pankreas, sehingga mengakibatkan kondisi hiperglikemia. Pembuatan hewan model DM tipe I menggunakan Streptozotocin dapat menyebabkan peningkatan kadar radikal bebas dan inflamasi pada organ hepar. Ekstrak albedo semangka merah (Citrullus vulgaris) memiliki kandungan sitrulin yang bersifat antioksidan dan anti inflamasi. Tujuan penelitian ini adalah untuk mengetahui pengaruh terapi ekstrak albedo semangka merah (Citrullus vugaris) terhadap sel Kupffer dan ekspresi IL-1β hepar tikus putih (Rattus norvegicus) model DM tipe I.Penelitian menggunakan hewan coba tikus putih (Rattus norvegicus) jantan strain wistar berumur 8-12 minggu berat badan rata-rata 150 gram, dibagi menjadi 5 kelompok dan setiap kelompok terdiri atas 4 ekor tikus putih (Rattus norvegicus), yaitu: kelompok K-, kelompok K+, dan 3 kelompok terapi (dosis 500 mg/kg BB, 1000 mg/kg BB, dan 1500 mg/kg BB). Terapi diberikan melalui sonde lambung selama 14 hari. Analisa data sel Kupffer dengan pewarnaan HE dan ekspresi IL-Iβ dengan pewarnaan imunohistokimia dilakukan secara deskriptif kualitatif dan kuantitatif. Analisa deskriptif kualitatif didasarkan atas pengamatan perubahan histopatologi yang menyertai keberadaan sel Kupffer dan ekspresi IL-1β hepar, analisa kuantitatif dilakukan menggunkan One Way ANOVA dilanjutkan dengan uji Tukey (p<0,05). Hasil penelitian menunjukkan dosis 1000 mg/kg BB dapat digunakan sebgai dosis terapi DM tipe I ditinjau dari penurunan jumlah sel Kupffer dan ekspresi Il-1β hepar tikus putih (Rattus norvegicus) model DM tipe I.

English Abstract

Type I diabetes mellitus (DM) is an endocrine system disorder characterized by decreased insulin production due to pancreatic β cell damage, resulting in hyperglycemia conditions. Making animal models of type I DM using Streptozotocin can cause increased levels of free radicals and inflammation in the liver organ. Red watermelon (Citrullus vulgaris) albedo extract has a content sitrulin as antioxidant and anti-inflammatory. The aim of this research is to know the therapy effect of red watermelon (Citrullus vugaris) albedo extract on liver Kupffer cell and expression of IL-1β white rat (Rattus norvegicus) type I diabetes models. Research using male white rats (Rattus norvegicus) wistar strain 8-12 weeks old, with average weight 150 grams, divided into 5 groups and each group consisted of 4 white rats (Rattus norvegicus): K-group, K + group, and 3 therapy gro ups (dose 500 mg/kg BW, 1000 mg/kg BW, and 1500 mg/kg BW). Treatment was given gastric sonde for 14 days. Analysis of Kupffer cell data with HE staining and IL-Iβ expression with immunohistochemical staining was done descriptively qualitative and quantitative. Qualitative descriptive analysis was based on observations of histopathological changes that accompanied the presence of liver Kupffer cells and IL-1β expression, a quantitative analysis was performed using One Way ANOVA followed by Tukey test (p <0.05). The results showed that 1000 mg/kg BW dose can be used as therapy dose of type I DM in terms of decreasing the liver number of Kupffer cells and IL-1β expression of white rat (Rattus norvegicus) of type I DM models.
