Abstract

Objektif : Tujuan dalam penelitian ini untuk mengetahui pengaruh ekstrak teh hijau (Camellia sinensis) berbagai dosis terhadap jumlah folikel Ovarium dan kadar 17β-estradiol serum pada tikus (Rattus norvegicus) yang dipapar monosodium glutamat. Metode : Penelitian ini merupakan penelitian true eksperimental dengan menggunakan pendekatan post test only control group design. Sampel yang digunakan adalah tikus putih betina (Rattus norvegicus) sejumlah 25 ekor yang terbagi menjadi 5 kelompok. Paparan monosodium glutamat diberikan dengan dosis 0,7 mg/grBB per oral dan ekstrak teh hijau berbagai dosis diberikan 2 jam setelahnya selama 30 hari setelah didapatkan tikus berada pada fase proestrus. Pemeriksaan jumlah folikel ovarium diukur menggunakan Dot slide mikroskop Olympus XC 10. Pemeriksaan kadar 17β-estradiol serum menggunakan kit Enzyme-Linked Immunosorbent Assay (ELISA) nomor katalog CSB-E05110r merk Cubasio. Data dianalisis menggunakan uji One Way Anova. Hasil : Berdasarkan hasil uji perbandingan berganda dengan uji LSD menunjukkan bahwa rerata jumlah folikel primer, sekunder dan de Graaf tertinggi dan folikel atresia terendah tampak pada kelompok perlakuan pemberian MSG dan ekstrak teh hijau dosis 1,4 mg/ekor/hari namun jumlah folikel primer, sekunder dan de Graaf menurun pada kelompok perlakuan pemberian MSG dan ekstrak teh hijau dosis 2,8 mg/ekor/hari dan meningkat pada folikel atresia. Nilai rerata kadar 17β-estradiol serum tertinggi tampak pada kelompok perlakuan pemberian MSG dan ekstrak teh hijau dosis 1,4 mg/ekor/hari (2.507±0.239) namun menurun pada kelompok perlakuan pemberian MSG dan ekstrak teh hijau dosis 2,8 mg/ekor/hari (2.204±0.147). Kesimpuan : Penelitian ini menunjukkan hasil pemberian ekstrak teh hijau berbagai dosis dapat meningkatkan jumlah folikel primer, sekunder, de Graaf dan menurunkan jumlah folikel atresia serta meningkatkan kadar 17β-estradiol serum pada tikus putih betina (Rattus norvegicus) yang dipapar monosodium glutamat

English Abstract

Objective : The aim of this study was to investigate the effect of green tea extract (Camellia sinensis) on various doses on Ovarian follicle count and serum 17β-estradiol levels in rats (Rattus norvegicus) exposed to monosodium glutamate. Method : This research is true experimental research using post test only control group design approach. The sample used was 25 female white rats (Rattus norvegicus) divided into 5 groups. Exposure to monosodium glutamate was administered at a dose of 0.7 mg / grBB orally and various green tea extracts were administered 2 hours later for 30 days after the rat found in the proestrus phase. Examination of the number of ovarian follicles was measured using Dot slide microscope Olympus XC 10. Serum 17β-estradiol concentration test using Enzyme-Linked Immunosorbent Assay (ELISA) catalog number CSB-E05110r Cubasio brand. Data were analyzed using One Way Anova test. Result : Based on the result of multiple comparison test with LSD test showed that the highest number of primary, secondary and de Graaf follicular follicles and lowest atresia follicle was seen in the treatment group of MSG and green tea extract dose 1.4 mg/day but the number of primary, secondary and de Graaf decreased in the treatment group of MSG and green tea extract dose 2.8 mg/day and increased in follicular atresia. The mean value of the highest serum 17β-estradiol level was seen in the treatment group of MSG and green tea extract dose 1.4 mg/day (2.507 ± 0.239) but decreased in the treatment group of MSG and green tea extract at 2.8 mg/day (2.204 ± 0.147). Conclusion: This study showed that the results of various green tea extract doses can increase the number of primary, secondary, de Graaf follicles and decrease the number of follicular atresia and increase the serum 17β-estradiol levels in female white rats (Rattus norvegicus) exposed to monosodium glutamate.
