Abstract
Penelitian mengenai interaksi METTL-10 (methyltransferase like-protein 10) dengan reseptornya sebagai marker komplikasi Diabetes Mellitus bertujuan untuk mengetahui perbedaan reaksi metilasi antara GAMT dan METTL-10 dengan enzim guanidinoasetat untuk menghasilkan produk Kreatin. Analisis dilakukan dengan melakukan docking melalui webserver PatchDock, FireDock dan pyDock dan divisualisasikan menggunakan Discovery Studio 2017 dan LigPlot+. Data yang didapatkan dari interaksi GAMT-GAA adalah energi Van der Waals (atraktif dan repulsif) sebesar -8,67 dan 0,58. Nilai energi kontak atom (ACE) sebesar -5,29 kkal/mol dengan nilai energi total ΔG0 adalah -20,79 kkal/mol dan nilai Ki sebesar 0,97. Untuk interaksi METTL-10-GAA, nilai energi Van der Waals (atraktif dan repulsif) sebesar -6.58 dan 1.13. Untuk energi kontak atom (ACE) yang dihasilkan sebesar -3,49 kkal/mol, sedangkan nilai energi bebas total ΔG0 adalah -10,42 kkal/mol dan Ki sebesar 0,983. Berdasarkan visualisasi dengan Discovery Studio 2017, interaksi berdasarkan sisi aktif GAMT-GAA dengan Kreatin memiliki kemiripan sebesar 90%, dan interaksi METTL-10-GAA untuk menghasilkan Kreatin sebesar 33,3%. Berdasarkan pengamatan/visualisasi data yang dihasilkan dan kemiripan sisi aktif METTL-10 dan GAMT dengan enzim guanidinoasetat, METTL-10 kemungkinan salah satu metil transferase yang terekspresi untuk menghasilkan Kreatin
English Abstract
This research concern about interaction between METTL-10 with its receptors as DM complications markers that aim to know the difference of methylation reaction about GAMT and METTL-10 with guanidinoacetate enzym to produces Creatine. Research using docking analysis by webserver PatchDock, FireDock and pyDock that visualized with Discovery Studio 2017 and LigPlot+. The data that can be obtained from GAMT-GAA interaction isVan der Waals (attractive and repulsive) as -8,67 and 0,58. For atomic contact energy (ACE) as -5,29 kkal/mol, total energy ΔG0 as -20,79 kkal/mol and inhibition constant (Ki) as 0,97. Interaction between METTL-10-GAA has Van der Waals (attractive and repulsive) as -6,58 and 1.13. For ACE reach -3,49 kkal/mol and Ki as 0,983. Based on visualization using Discovery Studio 2017, interaction between GAMT-GAA with Creatine has 90% similarity, and METTL-10-GAA has 33,3% similarity. Conclution from all of data that binding-site similarity between METTL-10 and GAMT with guanidinoacetate enzym, that probability METTL-10 is one of the methyltransferase that can be expressed to produces Creatine.
