Abstract

Minyak atsiri biji adas manis (Foeniculum vulgare Mill. var dulce) hingga saat ini belum menjadi komoditas utama walaupun memiliki potensi produksi. Isolasi minyak atsiri dapat dilakukan menggunakan metode distilasi uap. Salah satu faktor yang mempengaruhi proses distilasi adalah waktu distilasi. Waktu distilasi mempengaruhi kualitas dan profil komponen minyak atsiri. Komponen minyak atsiri mempengaruhi aktivitas antibakteri. Penelitian ini bertujuan untuk mengetahui pengaruh waktu distilasi terhadap rendemen, sifat fisik dan profil komponen minyak adas manis serta aktivitasnya sebagai antibakteri Staphylococcus aureus. Isolasi minyak atsiri biji adas manis dilakukan menggunakan distilasi uap selama 5, 7, dan 9 jam. Karakterisasi sifat fisik minyak atsiri dilakukan berdasarkan penentuan massa jenis, indeks bias, warna, aroma, dan wujud. Identifikasi profil komponen dilakukan menggunakan instrumen Kromatografi Gas-Spektrometer Massa (KG-SM). Uji antibakteri S. aureus minyak atsiri biji adas manis menggunakan metode difusi cakram. Minyak atsiri biji adas manis hasil penelitian ini mempunyai rendemen berturut-turut sebesar 1.1 %, 1.1 %, dan 1.19 %. Komponen yang teridentifikasi menggunakan analisis KG-SM berturut-turut sebanyak 7, 6 dan 11 senyawa. Lima komponen utama minyak adas manis yaitu anetol, fenkon, estragol, limonen dan α-pinen. Kadar senyawa anetol tertinggi (77,57 %) minyak biji adas manis diperoleh dari hasil distilasi uap 7 jam. Minyak atsiri yang diperoleh berwujud cair berwarna kuning jernih, beraroma spicy khas adas, mempunyai berat jenis 0.967-0.970 g/mL dan indeks bias 1.53-1.54. Diameter zona hambat minyak atsiri biji adas manis pada penelitian ini sebesar 8-11 mm, tergolong kategori resisten. Waktu distilasi mempengaruhi rendemen, profil komponen dan aktivitas antibakteri S.aureus minyak adas manis. Namun tidak mempengaruhi sifat fisik minyak atsiri biji adas manis.

English Abstract

Sweet fennel seed (Foeniculum vulgare Mill. var dulce) oils has not become a major commodity although it has potential production. Isolation of sweet fennel oil can be done by using steam distillation. One of the factor affecting distillation process is time distillation. Time distillation influence quality and component profile of essential oils. Components of essential oils influence its antibacterial activity. This study aims to know the time distillation influence on yield, physical properties and component profile of sweet fennel seeds oils also its antibacterial activity against Staphylococcus aureus. The isolation was done by using steam distillation for 5, 7, and 9 hours. Physical properties of sweet fennel seeds oils were determined by its state and appearance, color, odor, refractive index and specific gravity. Profile component was identified using Gas Chromatography-Mass Spectrometer (GC-MS). Antibacterial test of sweet fennel seeds oils was done using disk diffusion on S. aureus. The results showed that yield of sweet fennel seeds oils obtained by steam distillation respectively were 1.1 %, 1.1 % and 1.19 %. The identified components using GC-MS analysis are 7, 6 and 11 compounds, respectively. The five main components of sweet fennel oil is anethole, fenchone, estragole, limonene and α-pinene. The highest concentration of anethole (77.57 %) was obtained by steam distillation for 7 hours. Sweet fennel seeds oils obtained in this research is clear yellow liquid, has distinctive fennel spicy odor, with mass gravity of 0.967–0.970 g/mL and refractive index of 1.53–1.54. The antibacterial activity of sweet fennel seed oils against S. aureus is classified as resistance category (8-11 mm). Time distillation influence the yield, component profile and antibacterial activity of sweet fennel seeds oils against S. aureus, but it does not influence the physical properties of sweet fennel seeds oils.
