Abstract

Penelitian ini bertujuan untuk mengetahui pemberian ekstrak Etanol Daun Tapak Liman (Elephantopus scaber) terhadap Tingkat Nekrosis Sel Ginjal dan Hati Mencit Model Kanker Payudara. Penelitian ini menggunakan 27 ekor mencit betina BALB/c yang berumur lima minggu. Hewan coba dibagi menjadi tiga kelompok dan tiga waktu pembedahan dengan tiga ekor mencit per kelompok. Sebelum pemberian ekstrak dilakukan permodelan kanker payudara dengan induksi senyawa karsinogen DMBA (7,12- dimethylbenz(a)anthracene). Kemudian, ekstrak tapak liman diberikan secara oral selama 7 hari, 14 hari dan 21 hari dengan dosis 0 mg ekstrak kg/BB untuk kontrol dan dosis 50 mg ekstrak /kg BB sebagai perlakuan pertama. Setiap pemberian perlakuan 7 hari, 14 hari dan 21 hari, dibedah dan diambil organ hepar dan ginjal untuk dibuat preparat histologi dengan metode parafin dan pewarnaan Hematoxylin-Eosin. Rata-rata sel nekrosis pada jaringan hepar dan ginjal diamati dan dihitung pada lima bidang pandang menggunakan mikroskop perbesaran 400x. Hasil penelitian ini yaitu ekstrak etanol daun Elephantopus scaber mempengaruhi nekrosis sel hepatosit pada hati dengan lama pemberian yang paling baik selama 7 hari dengan rata-rata sel nekrosisnya 26,28% dan menurunkan sel nekrosis pada sel epitel tubulus ginjal dan walaupun tidak berbeda nyata lama pemberian paling baik selama 14 hari adalah 29,12%. Data yang diperoleh dianalisis menggunakan ANOVA (p  0.05) dengan program SPSS.

English Abstract

This study aims to determine the adduction of etanol leaf extract of Tapak Liman (Elephantopus scaber) to Kidney and Liver Cell Necrosis Level on Mice Breast Cancer Model. This study used 27 female mice BALB /c of 5 weeks old. Animals were divided into three groups and three surgeries with three mice per group. Before administering extracts, were performed breast cancer modeling by induction of DMBA carcinogen compound (7,12-dimethylbenz (a) anthracene). Then, the extract administered orally for 7 days, 14 days and 21 days with a dosage of 0 mg of extract kg / BB for control and dose of 50 mg extract / kg BW as treatment 1. Each treatment 7 days, 14 days and 21 days , isolated liver and kidney organs to make histology preparations with paraffin and Hematoxylin-Eosin staining methods. The average necrosis cell of hepatic and renal tissue was observed and calculated on five field of view using a 400 x magnification microscope. The results of this research is the extract of ethanol leaves Elephantopus scaber affect the hepatocyte cell necrosis in the liver with the best duration of 7 days with the average cell necrosis is 26.28% and decrease the necrosis cells in ritual tubule epithelial cells and although not significantly different length of administration Best for 14 days is 29.12%. The data obtained were analyzed using ANOVA (p  0.05) with SPSS program. The results showed that the tread liman extract affected the number of hepatocytes and necrotic renal tubular cells although not significantly.
