Abstract
Traumatic Brain Injury merupakan salah satu kerusakan fungsi otak yang dapat menyebabkan gangguan neurologis pada otak. Gangguan ini dapat disebabkan benturan benda keras di kepala sehingga mengakibatkan peningkatan tekanan intrakarnial dan timbulnya radikal bebas. Ekstrak kulit manggis (Garcinia mangostana L.) diketahui mengandung senyawa xanton yang bekerja sebagai antioksidan dan berfungsi sebagai antiinflamasi. Penelitian ini bertujuan untuk mengetahui profil protein serum dan ekspresi SOD pada tikus (Rattus norvegicus) model traumatic brain injury. Penelitian ini menggunakan hewan coba tikus yang dikelompokkan menjadi 4 perlakuan yaitu kontrol negatif, kontrol positif (model TBI), kelompok TBI dengan terapi ekstrak kulit manggis dosis 0,5 mL/hari selama 5 hari dan kontrol TBI dengan terapi minocycline dosis 0,5 mL/hari selama 5 hari. Profil protein dianalisis menggunakan metode elektroforesis SDS-PAGE dan ekspresi SOD pada otak dianalisis secara deskriptif sel-sel yang terekspresi. Hasil penelitian menunjukkan bahwa terapi ekstrak kulit manggis terbentuk pita protein baru dengan BM 79,034 kDa dan meningkatkan ekspresi SOD pada jaringan otak tikus sebesar 78%. Hasil analisa statistik ekspresi SOD menunjukkan bahwa terdapat perbedaan yang signifikan antar kelompok perlakuan (p<0,01). Penelitian ini dapat disimpulkan bahwa terapi ekstrak kulit manggis dapat digunakan untuk terapi pasca cedera otak.
English Abstract
Traumatic Brain Injury is one of the brain damage that can cause neurological disorders in the brain. This disorder can be caused by a collision of hard objects in the head resulting in increased intracarnial pressure and the emergence of free radicals. The mangosteen skin extract (Garcinia mangostana L.) is known to contain xanton compounds that act as antioxidants and act as anti-inflammatory. The aim of this study was to determine the effect of mangosteen pericarp extract therapy on serum protein profile and increased expression of SOD on traumatic brain injury rats (Rattus norvegicus). This study used the experimental animals grouped into 4 groups treatment: negative group, positive group(TBI), TBI group with mangosteen pericarp extract therapy dose 0,5 mL/day for 5 days and TBI group with minocycline therapy dose 0,5 mL/day for 5 days. To find serum protein profiles using SDS-PAGE method and SOD expression in brain was analyzed descriptively by expressed cells. The results showed that mangosteen pericarp therapy formed a new protein band with 79.034 kDa of BM. Treatment of mangosteen pericarp extract able to increase SOD expression up to 78%. The result of statistical analysis of protein profile and SOD expression showed that there were significantly differences between treatment groups(p<0,01). It can be concluded that mangosteen pericarp extract therapy can be used therapy after brain injury.
