Abstract
Diare merupakan penyakit yang ditandai dengan bertambahnya frekuensi buang air besar dengan disertai perubahan bentuk dan konsistensi tinja pada penderita. Enterophatogenic Eschercia coli (EPEC), merupakan strain pertama dari E. coli yang diidentifikasi sebagai penyebab diare patogenik pada pasien bayi dan anak-anak yang menyebabkan kerusakan pada daerah usus halus. Bakteri Asam Laktat (BAL) yang terkandung dalam Water Soluble Extract (WSE) yoghurt susu kambing dapat digunakan sebagai preventif diare karena adanya kandungan bioaktif peptida yang berperan sebagai antibakterial. Tujuan penelitian ini untuk mengetahui pengaruh pemberian preventif WSE yoghurt susu kambing sebagai antidiare pada tikus putih yang diinfeksi EPEC terhadap gambaran histopatologi dan ekspresi TNF - α Jejunum. Penelitian ini menggunakan lima kelompok perlakuan yaitu kontrol positif (EPEC), kontrol negatif, dan kelompok prevetif WSE yoghurt susu kambing dosis 300 mg/Kg BB, 600 mg/Kg BB, dan 900 mg/Kg BB per oral selama 14 hari. Parameter penelitian ini adalah gambarang histopatologi kerusakan vili jejunum yang dianalisis secara deskriptif kualitatif dan pengukuran ekspresi TNF - α jejunum yang dideskripsikan dengan software Immunoratio kemudian dianalisis menggunakan uji ANOVA dilanjutkan dengan uji BNJ dengan α = 0,05. Hasil penelitian menunjukkan bahwa preventif WSE yoghurt susu kambing secara signifikan (α = 0,05) mampu menghambat peningkatan ekspresi TNF - α dan mempertahan gambaran histologi Jejunum seperti terhambatnya erosi pada vili jejunum. Dosis 900 mg/Kg BB merupakan dosis efektif dengan penurunan ekspresi TNF - α sebesar 49,78 %. Kesimpulan dari penelitian ini pemberian preventif WSE yoghurt susu kambing dapat digunaklan sebagai alternatif pencegahan diare akibat infeksi EPEC.
English Abstract
Diarrhea is a disease characterized by increasing frequency of defecation with changes in the shape and consistency of feces in the patient. Enterophatogenic Eschercia coli (EPEC), is the first strain from E.coli identified as a cause of pathogenic diarrhea in baby and children who caused damage to the small intestine. Lactid Acid Bacteria (LAB) contain in Water Soluble Extract Goat Milk Yoghurt can be used as a diarrhea preventive due to contains bioactive peptide that act as an antibacterials. The purpose of this study was to investigate the effect of WSE Goat Milk Yoghurt for preventing diarrhea in Rattus Norvegicus that infected of EPEC based on histopathology and expression TNF- α of Jejenum. This research were using 5 groups, such as : the group of positive diarrhea (EPEC), the group of negative control, and group that was gaven WSE Goat Milk Yoghurt about 300mg/kgBB, 600mg/kg BB and 900mg/kg BB PO in 14days. The parameter being measured are the damage of jejenum tisuue that analyzed by descriptively qualitativeand measurment of TNF- α expression that discribe with Immunoratio software and than analyzed by statistically using one-way ANOVA α = 0,05. The result show that Goat Milk Yoghurt as preventiv can inhibit the increased of TNF- α and maintaince histological of Jejenum. Effective dose in this research is 900mg/kg BB, because it can decreased the expression TNF- α amount 49,78%. The conclusion of this research are, suplementation of WSE Goat Milk Yoghurt can use for alternative to prevent diarrhea caused by EPEC infection.
