Abstract
Pemberian antibiotik merupakan salah satu pilihan untuk mengobati gastroenteritis akibat infeksi Escherichia coli, tetapi dapat menimbulkan resistensi jika tidak digunakan secara teratur, maka perlu dicarikan dan dilakukan penelitian pengobatan alternatif salah satunya seperti ekstrak etanol daun sawo. Daun sawo (Manilkara zapota L.) mengandung senyawa flavonoid dan tannin yang berperan sebagai antibakteri dan antioksidan. Penelitian ini bertujuan untuk mengetahui efek terapi ekstrak etanol daun sawo (Manilkara zapota L.) terhadap gastroenteritis pada tikus putih (Rattus norvegicus) jantan yang diinduksi Escherichia coli dilihat dari ekspresi IL-1B dan histopatologi jejunum. Rancangan penelitian ini bersifat eksperimental dengan post test control only menggunakan rancangan acak lengkap (RAL). Tikus yang dipakai dalam penelitian ini adalah tikus putih (Rattus norvegicus) jantan berumur 5-6 minggu yang dibagi menjadi 5 kelompok, yaitu: kelompok A (kontrol negatif), kelompok B (kontrol positif diinduksi E. coli 106 CFU/ml), kelompok C, D dan E (kelompok yang diinduksi E. coli 106 CFU/ml dan terapi ekstrak etanol daun sawo selama 8 hari dengan dosis secara berurutan 200 mg/kg BB, 300 mg/kg BB dan 400 mg/kg BB). Parameter dalam penelitian ini adalah ekspresi IL-1B dengan metode imunohistokimia yang dianalisis dengan one way ANOVA dengan tingkat kepercayaan =0.05 dan histopatologi jejunum dianalisis secara deskriptif. Kesimpulan dari penelitian ini adalah ekstrak etanol daun sawo (Manilkara zapota L.) dapat digunakan sebagai terapi alternatif gastroenteritis dengan dosis 400 mg/kg BB merupakan dosis terbaik yang dapat menurunkan ekspresi IL-1B paling tinggi hingga mencapai 25,90% dan dapat memperbaiki kerusakan vili jejunum.
English Abstract
Antibiotic is one option for treating gastroenteritis due to Escherichia coli, but can lead to resistance if not used properly, then alternative treatments are needed such as ethanol extract of sawo manila leaves (Manilkara zapota L.). The sawo manila leaves (Manilkara zapota L.) contain flavonoid and tannin compounds that act as antibacterial and antioxidant. This research aims to know the therapeutic effects of ethanol extracts of sawo manila leaves (Manilkara zapota L.) against gastroenteritis in male white rats (Rattus norvegicus) induced Escherichia coli as seen from IL-1B expression and jejunum histopathology. The design of this research are experimental with post test control only using random design complete (RAL). These rats are used in this study is the male white rat (Rattus norvegicus) was 5-6 weeks are divided into 5 groups: group A (negative control), group B (positif control that induced Escherichia coli 106 CFU/ml) group C, D and E (the group that induced Escherichia coli 106 CFU/ml and ethanol extracts therapy of sawo manila leaves for 8 days with dose sequentially 200 mg/kg BW, 300 mg/kg BW and 400 mg/kg BW. The parameters in this study are the IL-1B expression with immunohistochemistry that analyzed by one way ANOVA with a confidence level is =0.05 and jejunum histopathology analyzed was descriptively. The conclution of this study is ethanol extract of sawo manila leaves could use as gastroenteritis alternative therapy wich a dose of 400 mg/kg WB was the most effective that could decreas IL-1B expression most high-reaching 25,90% and could repair damage on jejunum villi.
