Abstract
Latar Belakang : Monosodium Glutamat (MSG) adalah garam natrium dari asam glutamat. Asam glutamat merupakan asam amino non esensial yang nantinya dikonversi menjadi glutamat di dalam tubuh. Fungsi utama glutamat sebagai komunikasi antar sel, dan juga dapat mengendalikan GnRH. Mengkonsumsi MSG yang berlebihan akan menyebabkan jumlah glutamat dalam plasma darah meningkat sehingga terjadi over aktivasi reseptor glutamat yang memicu terjadinya stres oksidatif. Dampak dari stres oksidatif pada hipotalamus mengakibatkan gangguan pada area pituitary adrenal axis sehingga hipofise anterior terganggu dalam mensekresi FSH dan LH yang dapat mempengaruhi perkembangan ukuran folikel. Sedangkan pada organ reproduksi, glutamat yang berlebihan menyebabkan kerusakan jaringan sel. Tujuan : untuk mengetahui pengaruh paparan MSG dan pemberian ekstrak etanol teh hijau terhadap pencegahan penurunan kadar FSH, pencegahan peningkatan kadar Malondialdehyde (MDA) ovarium dan diamater folikel antral. Bahan dan metode : 25 ekor tikus putih betina (Rattus norvegicus) Galur Wistar dibagi 5 kelompok, masing-masing 5 ekor; K(-), K(+) : MSG dosis 0,7 mg/gBB, PI : MSG dosis 0,7 mg/gBB + ekstrak etanol teh hijau 0,7 mg, PII : MSG dosis 0,7 mg/gBB + ekstrak etanol teh hijau 1,4 mg dan PIII : MSG dosis 0,7 mg/gBB + ekstrak etanol teh hijau 2,8 mg. Perlakuan dilakukan selama 30 hari. Kadar FSH dianalisis melalui elisa essay, kadar MDA dianalisis melalui spektofotometri dan diameter folikel antral dianalisis melalui pewarnaan hematoksilin eosin kemudian diukur dengan software olyvia dan dotslide. Hasil : analisis statistik dengan one way anova, kadar FSH dan MDA ovarium diperoleh p value < 0,05 menunjukkan bahwa terdapat perbedaan yang signifikan kadar FSH dan MDA ovarium antara kelompok perlakuan dan kontrol akibat induksi MSG dan pemberian ekstrak teh hijau. Sedangkan diameter folikel antral dengan uji one way anova diperoleh p value > 0,05, yaitu tidak adanya perbedaan ukuran diameter folikel antral antara kelompok perlakuan dan kontrol akibat induksi MSG dan pemberian ekstrak etanol teh hijau. Kesimpulan : pemberian ekstrak etanol teh hijau dapat mencegah penurunan kadar FSH, dan mencegah peningkatan kadar MDA ovarium pada tikus betina yang terpapar monosodium glutamat. Tetapi hipotesis tentang pemberian ekstrak etanol teh hijau dapat mencegah penurunan diamater folikel antral tidak terbukti.
English Abstract
Background: Monosodium Glutamate (MSG) is a sodium salt of glutamic acid. Glutamic acid is a non-essential amino acid that is later converted into glutamate in the body. The main function of glutamate is as a communicator between cells, and also control GnRH. Consumption of MSG causes the amount of glutamate in the blood plasma increase resulting the over activation of glutamate receptors that triggers the oxidative stress. The damage of hypothalamus results the disruption of the pituitary adrenal axis area so that the anterior hypophysis is hampered in secreting FSH and LH which may affect the development of follicle size. While in the reproductive organs, excessive glutamate causes the cell tissue damage. Objective: This study aims to determine the effect of MSG exposure and green tea ethanol extract on the prevention of decreased levels of FSH, prevention of the increasing levels of ovarian Malondialdehyde (MDA) and antral follicle diameter. Materials and methods: 25 female rats (Rattus norvegicus) wistar strain divided by 5 groups, each of 5 mice; K (-), K (+): MSG dose 0.7 mg /gBB, PI: MSG dose 0.7mg /gBB + green tea ethanol extract 0.7 mg, PII: MSG dose 0.7mg /gBB + green tea ethanol extract 1,4 mg and PIII: MSG dose 0,7mg /gBB + green tea ethanol extract 2,8 mg. Treatment for 30 days. FSH levels were analyzed by elite essay, MDA levels analyzed by spektofotometri and antral follicle diameter were analyzed by eosin hematoxylin staining and then measured with olyvia and dotslide software. Results: statistical analysis by one way anova, FSH and MDA ovaries obtained p value <0,05 indicated that there was significant difference of FSH and MDA ovarian level between treatment and control group induced by MSG and green tea extract. While the diameter of antral follicle by one way anova test obtained p value> 0,05, there is no difference of antral follicle diameter size between treatment and control group due to MSG induction and green tea extract. Conclusion: MSG exposure and green tea ethanol extract can increase FSH levels and decrease MDA ovary level of female rat, but hypothesis about MSG exposure and green tea ethanol extract can increase antral follicle diameter is not proven.
