Abstract

Latar Belakang: Bakteri Lactobacillus acidophilus merupakan bakteri gram prositif yang umum ditemukan di rongga mulut dan berkaitan erat dengan proses perkembangan karies gigi. Kasein susu kambing peranakan etawa memiliki kandungan peptida bioaktif yang dapat menghambat aktivitas bakteri gram positif dan gram negatif. Tujuan: Mengetahui efektivitas kasein susu kambing peranakan etawa sebagai antibakteri terhadap Lactobacillus acidophilus. Metode: Metode yang digunakan yaitu metode dilusi untuk menentukan KHM. KBM dilakukan menggunakan Total Plate Count. Konsentrasi pasta kasein yang digunakan yaitu 20%, 10%, 5%, 2,5%, kontrol positif chx 0,2%, kontrol negatif aquades, dan kontrol pembanding CPP-ACP. Hasil: KHM pada penelitian ini tidak dapat ditentukan karena kekeruhan yang diakibatkan oleh bahan uji. Hasil hitung koloni diperoleh rata-rata jumlah koloni sebanyak 388 koloni pada konsentrasi 20%, 425 koloni pada konsentrasi 10%, 271 koloni pada konsentrasi 5%, 624 koloni pada konsentrasi 2,5%, 0 koloni pada kontrol positif chx 0,2%, 300 koloni pada kontrol negatif aquades, dan 213 koloni pada kontrol pembanding CPP-ACP, sehingga KBM pasta kasein tidak dapat ditentukan. Kesimpulan: Pasta kasein susu kambing peranakan etawa tidak efektif sebagai antibakteri terhadap Lactobacillus acidophilus.

English Abstract

Background: Lactobacillus acidophilus bacteria are gram-positive bacteria that are commonly found in the oral cavity and are closely related to the development of dental caries. Casein of PE goat milk contains bioactive peptides that can inhibit the activity of grampositive and gram-negative bacteria. Objective: To determine the effectiveness of etawa goat milk casein as an antibacterial against Lactobacillus acidophilus. Methods: The method used was dilution method to determine MIC. MBC is carried out using Total Plate Count. The casein paste concentrations used were 20%, 10%, 5%, 2.5%, positive control chx 0,2%, negative control aquades, and comparison control CPP-ACP. Results: MIC in this study could not be determined because the turbidity caused by the test material. The results of the colony count showed that the average number of colonies was 388 colonies at a concentration of 20%, 425 colonies at a concentration of 10%, 271 colonies at a concentration of 5%, 624 colonies at a concentration of 2.5%, 0 colonies at the positive control chx 0.2% , 300 colonies in the negative control of distilled water, and 213 colonies in the CPP-ACP comparison control, so the MBC of casein paste could not be determined. Conclusion: Etawa goat milk casein paste is not effective as an antibacterial against Lactobacillus acidophilus.
