Abstract

NAFLD (Non-Alcoholic Fatty Liver Disease) adalah gangguan hati yang mana terjadi infiltrasi lemak, terutama trigliserida pada sel hepatosit hingga berat organ hati meningkat lebih dari 5%. Komponen sindrom metabolik yang sering ditemukan adalah peningkatan kadar gula darah yang tidak terkontrol atau bisa disebut dengan hiperglikemia. Penatalaksanaan NAFLD meliputi perubahan gaya hidup, farmakoterapi dengan metformin, atau pendekatan lain seperti probiotik. Tujuan dilakukannya penelitian ini adalah untuk mengetahui pengaruh pemberian kombinasi metformin dan lactobacillus plantarum DAD - 13 terhadap kadar gula darah dan skeletal muscle mass index pada tikus model NAFLD. Metode yang digunakan dalam penelitian ini meruapada penelitian eksperimental murni (true experimental) dilakukan secara in vivo dengan posttest only group design. Hewan coba dibagi menjadi beberapa kelompok, yaitu diet normal (DN) sebagaikelompok kontrol, western diet (WD), dan Kelompok Metformin dan L. plantarum Dad-13 (KMP). Pengukuran kadar gula darah melalui ekor mendapati hasil bahwa kelompok western diet dengan pemberian kombinasi metformin dan Lactobacillus plantarum DAD – 13 memiliki kadar gula darah paling tinggi diikuti dengan kelompok diet normal dan pada kadar gula darah terendah terdapat kelompok western diet. Begitupula pada hasil pengukuran darah jantung dan GOD-PAP, kelompok western diet dengan pemberian kombinasi metformin dan Lactobacillus plantarum DAD – 13 memiliki kadar gula darah paling tinggi. Kesimpulan yang didapat adalah kombinasi metformin dan Lactobacillus plantarum DAD – 13 memberikan efek peningkatan rata rata kadar gula darah dan penurunan pada rata rata Skeleta muscle mass index.

English Abstract

NAFLD (Non-Alcoholic Fatty Liver Disease) is a liver disorder in which fat infiltration occurs, especially triglycerides in hepatocyte cells until the weight of the liver increases by more than 5%. A component of metabolic syndrome that is often found is an uncontrolled increase in blood sugar levels or what can be called hyperglycemia. Management of NAFLD includes lifestyle changes, pharmacotherapy with metformin, or other approaches such as probiotics. The aim of this experiment is to determine the effect of administering a combination of metformin and lactobacillus plantarum DAD - 13 on blood sugar levels and skeletal muscle mass index in NAFLD model mice. This experiment is true experimental research (true experimental) was carried out in vivo with a posttest only group design. The experimental animals were divided into several groups, namely normal diet (DN) as the control group, western diet (WD), and the Metformin and L. plantarum Dad-13 (KMP) group. Measuring blood sugar levels through the tail showed that the western diet group given a combination of metformin and Lactobacillus plantarum DAD – 13 had the highest blood sugar levels followed by the normal diet group and the western diet group had the lowest blood sugar levels. Likewise, in the results of heart blood and GOD-PAP measurements, the western diet group given a combination of metformin and Lactobacillus plantarum DAD – 13 had the highest blood sugar levels. In conclusion the combination of metformin dan Lactobacillus plantarum DAD – 13 has the effect of increasing the avarage blood sugar level and decreasing the avarage skeletal muscle mass index
