Aktivitas Antimalaria Ekstrak Etanol Batang Kayu Songga (Strychnos lucida R. Br.) Terhadap Plasmodium berghei ANKA Secara In Vivo.

Abstract

Malaria menjadi salah satu penyakit dengan resiko penularan dan keparahan yang tinggi. Tanaman songga dikenal sebagai obat demam dan malaria di wilayah Nusa Tenggara Barat (NTB) dan Bali. Kayu songga dimaserasi menggunakan etanol 80%. Kemudian dilakukan preparasi bahan uji menggunakan larutan CMC Na 0,5% sehingga didapatkan tiga macam dosis larutan uji antara lain 100, 10, dan 1 mg/kg BB mencit. Terdapat empat parameter yang diamati, (1.) Persentase parasitemia (2.) Persentase pertumbuhan parasitemia (3.) Persentase hambatan pertumbuhan parasitemia (4.) Nilai survival time mencit. Plasmodium berghei ANKA diinfeksikan secara intraperitoneal pada mencit jantan strain Balb/C berusia 2 bulan dengan berat badan 20-30 gram. Klorokuin dengan dosis 10 mg/kg BB mencit digunakan sebagai kontrol negatif dan CMC Na 0,5% sebagai kontrol positif. Pengujian dilakukan berdasarkan Peter’s Test, yaitu pemberian larutan uji selama 4 hari dan pembuatan hapusan darah tepi selama 7 hari untuk pengamatan terhadap persentase parasitemia dilanjutkan dengan pengamatan survival time hingga 28 hari. Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol kayu songga tidak memberikan perbedaan yang signifikan terhadap persentase parasitemia dan nilai survival time mencit dengan nilai P>0,05. Namun hasil menunjukkan adanya perbedaan yang signifikan terhadap persentase pertumbuhan parasitemia dengan nilai P<0,05. Selain itu pemberian ekstrak etanol kayu songga memiliki kemampuan dalam menghambat pertumbuhan Plasmodium berghei ANKA dibandingkan dengan kelompok kontrol positif dengan persen hambatan sebesar 43,7% (dosis 100 mg/kgBB), 33,5%(dosis 10 mg/kgBB) dan 33%(dosis 1 mg/kgBB). Hasil analisis profil metabolit sekunder menunjukkan kandungan senyawa dengan kandungan tertinggi adalah kelompok alkaloid dan fenol dengan persen area masing-masing 45,98% dan 22,18%.

English Abstract

Malaria is a disease with a high risk of transmission and severity. Songga plant is a medicine for fever and malaria in West Nusa Tenggara (NTB) and Bali. Songga wood had macerated using 80% ethanol. The sample preparations had conducted with 0.5% Na CMC solution. So that three different doses of the test solution were obtained, including 100, 10, and 1 mg/kg BW of mice. There are four parameters, (1.) The percentage of parasitemia (2.) The growth percentage of parasitemia (3.) The growth inhibition of parasitemia (4.) Survival time of mice. Plasmodium berghei ANKA was injected intraperitoneally in 2-month-old male mice of Balb/C strain with a body weight of 20-30 grams. Chloroquine 10 mg/kg BW in mice use as a negative control and 0.5% CMC Na as a positive control. The test was carried out based on Peter's Test, namely giving the test solution for four days and making blood smears for seven days to observe the percentage of parasitemia, followed by observation of survival time up to 28 days. The results showed that the administration of ethanol extract of songga wood did not give a significant difference to the percentage of parasitemia and the survival time of mice with a P>0.05. However, the results showed a significant difference in the growth percentage with a P<0.05. Besides that, the administration of ethanol extract of songga wood could inhibit the growth of Plasmodium berghei compared to the positive control group with a percentage of inhibition of 43.7% (dose of 100 mg/kg BW), 33.5% (dose of 10 mg/kg BW) and 33% (dose of 1 mg/kg BW). The results of the metabolite profile analysis showed that the compounds with the highest content were alkaloids and phenol groups with a percentage of the area of 45.98% and 22.18%, respectively.
