Abstract

Methicillin-Resistant Staphylococcus aureus (MRSA) menjadi masalah kesehatan dunia akibat peningkatan angka resistensi antibiotik yang terus meningkat. MRSA merupakan salah satu patogen penyebab infeksi pada fasilitas pelayanan kesehatan. Peningkatan laju resistensi ini jika tanpa diimbangi dengan penemuan baru menimbulkan kekhawatiran. Untuk mengatasinya dibutuhkan suatu inovasi agen antibakteri yang murah, mudah didapatkan, serta melimpah. Tanaman jeruk purut (Citrus hystrix) merupakan tanaman asli Indonesia dan tersebar luas di seluruh dunia. Tanaman jeruk purut, khususnya daunnya, sering digunakan sebagai obat tradisional dan bumbu masakan. Penelitian ini didesain sebagai penelitian eksperimental (true experimental) untuk menguji aktivitas antibakterial terhadap koloni MRSA setelah diberikan ekstrak Daun Jeruk Purut menggunakan metode difusi sumuran. Kelompok perlakuan terdiri dari kontrol negatif menggunakan aquades, ekstrak daun jeruk purut konsentrasi 75%, 80%, 85%, 90%, 95%, 100%, serta kontrol positif menggunakan Vancomycin 20 μg/ml Rerata diameter zona hambat ekstrak Daun Jeruk Purut secara berturut-turut menunjukkan nilai 8, 9.5, 9.6, 10, 10.3, dan 10.9mm. Hasil penelitian menunjukkan adanya perbedaan signifikan dari seluruh kelompok perlakuan konsentrasi ekstrak terhadap kontrol negatif. Sedangkan uji korelasi Spearman menunjukkan korelasi positif sangat kuat. Berdasarkan data penelitian, dapat disimpulkan bahwa ekstrak Daun Jeruk Purut (Citrus hystrix) memiliki efek terhadap pertumbuhan bakteri MRSA secara in vitro dengan metode difusi sumuran.

English Abstract

Methicillin-Resistant Staphylococcus aureus (MRSA) is a global health problem due to the increasing prevalence of antibiotic resistance. MRSA is one of the pathogens that cause infection in health care settings. The imbalance between resistance rate and discovery of new antibiotic raises concerns. It urge to find an innovative antibacterial agent that is cheap, easy to obtain, and abundant. Lime (Citrus hystrix) is a native plant of Indonesia and is widely distributed throughout the world. Jeruk Purut, especially the leaves, are often used as traditional medicine and cooking spices. This research was designed as an experimental study (true experimental) by observing the antibacterial activity of the extract of Jeruk Purut leaves against MRSA colonies using the well diffusion method. The treatment group consisted of negative control using aquadest, Purut Jeruk Leaf Extract with concentrations of 75%, 80%, 85%, 90%, 95%, 100%, and positive control using Vancomycin 20 g/mL. The average diameter of the inhibitory zone of Jeruk Purut leaf extract showed values of 8, 9.5, 9.6, 10, 10.3, and 10.9mm, respectively. The results showed that there was a significant difference from all extract treatment groups to the negative control. Spearman correlation test showed a positive correlation of 0.962 (very strong). As conclusion, the extract of lime leaves (Citrus hystrix) has an effect on the growth of MRSA bacteria in vitro by the well diffusion method
