Abstract

RINGKASAN Berdasarkan data dari Badan Pusat Statistik (2021) Pada tahun 2021 Indonesia mempunyai total lahan perkebunan kelapa sawit seluas 14.663,60 juta Ha. Hasil panen yang diperoleh mencapai 46.223,30 juta ton (Badan Pusat Statistik, 2021). Tingginya produksi CPO memberikan dampak buruk bagi lingkungan dikarenakan terdapat limbah yang dihasilkan dari proses produksi. Berdasarkan hasil penelitian yang dilakukan Wibowo et al. (2019), 23% dari total panen kelapa sawit berupa tandan kosong yang tidak dapat dimanfaatkan. komposisi serat penyusun TKKS diantaranya lignin (22,6%), selulosa (45,8) dan hemiselulosa (31,8%). Paenibacillus sp termasuk agent mikrobiologi yang digunakan dalam pretreatment biologi, dimana bakteri ini dapat menghasilkan enzim ligninolitik berupa enzim MnP dan LiP yang mampu mendegradasi ikatan kovalen lignin. Diduga dengan adanya penambahan induser CuSO4 serta variasi suhu inubasi dapat mampu meningkatkan degradasi senyawa lignoselulosa terkhusus pada senyawa polimer lignin. Penelitian ini bertujuan untuk menganalisis pengaruh penggunaan inducer CuSO4 dan pengaruh suhu inkubasi terhadap proses pre-treatment limbah tandan kosong kelapa sawit serta mengevaluasi penambahan CuSO4 sebagai sumber nutrisi pertumbuhan bakteri Paenibacillus sp dan delignifikasi limbah tandan kosong kelapa sawit. Pada penelitian ini, pretreatment dilakukan dengan menggunakan bakteri Paenibacillus sp sebagai mikroorganisme pendegradasi lignin terhadap biomassa lignoselulosa TKKS. Pengujian yang dilakukan pada penelitian meliputi uji Total Soluble Phenol (TSP), uji Total Gula Reduksi (TGR), Uji Kadar Lignoselulosa; Uji Kadar Air; Uji weight loss; uji pH. Rancangan penelitian yang digunakan adalah Rancangan Acak Kelompok (RAK) menggunakan 2 faktor. Faktor konsentrasi induser CuSO4 (K) dengan 3 (tiga) level konsentrasi 0,2 mM; 0,5 mM; dan 0,8 mM. Faktor variasi suhu inkubasi (S) dengan tiga level suhu 250C, 300C, dan 350C. Terdapat 9 kombinasi sampel berbeda antara lain K1S1, K1S2, K1S3, K2S1, K2S2, K2S3, K3S1, K3S2, K3S3. Setiap perlakuan dilakukan pengulangan sebanyak 3 (tiga) kali. Hasil dari pengujian akan dianalisis menggunakan ANOVA dan uji DMRT. Hasil dari penelitian yang dilakukan yaitu Nilai TGR tertinggi berada pada sampel dengan penambahan induser CuSO4 0,8mM sebesar 10,645 mg/g. Nilai TSP tertinggi yaitu sampel dengan penambahan induser CuSO4 0,5mM sebesar 0,0343mg/g. pH Seluruh sampel mengalami penurunan (mendekati asam) dan nilai removal lignin paling tinggi sebesar 23,108% sampel dengan penambahan induser CuSO4 0,5mM. Induser CuSO4 memberikan pengaruh terhadap delignifikasi.

English Abstract

SUMMARY Based on data from the Central Statistics Agency (2021), in 2021 Indonesia has a total area of 14,663.60 million hectares of oil palm plantations. The harvest obtained reached 46,223.30 million tons (Central Statistics Agency, 2021). The high production of CPO has a negative impact on the environment because there is waste generated from the production process. Based on the results of research conducted by Wibowo et al. (2019), 23% of the total oil palm harvest is in the form of empty bunches that cannot be utilized. The composition of EFB constituent fibers includes lignin (22.6%), cellulose (45.8) and hemicellulose (31.8%). Paenibacillus sp is a microbiological agent used in biological pretreatment, where these bacteria can produce ligninolytic enzymes in the form of MnP and LiP enzymes that are able to degrade lignin covalent bonds. It is suspected that the addition of CuSO4 inducers and variations in inubation temperature can be able to increase the degradation of lignocellulose compounds, especially in lignin polymer compounds. This study aims to analyze the effect of the use of CuSO4 inducers and the effect of incubation temperature on the pre-treatment process of empty oil palm bunch waste and evaluate the addition of CuSO4 as a source of nutrients for the growth of Paenibacillus sp bacteria and delignification of empty oil palm bunch waste. This study aims to analyze the effect of the use of CuSO4 inducers and the effect of incubation temperature on the pre-treatment process of empty oil palm bunch waste and evaluate the addition of CuSO4 as a source of nutrients for the growth of Paenibacillus sp bacteria and delignification of empty oil palm bunch waste. In this study, pretreatment was carried out using Paenibacillus sp bacteria as lignin degrading microorganisms against EFB lignocellulose biomass. Tests conducted in the study include Total Soluble Phenol (TSP) test, Total Sugar Reduction (TGR) test, Lignocellulose Content Test; moisture content test; Test weight loss; pH test. The research design used was Group Randomized Design (RAK) using 2 factors. CuSO4 (K) inducer concentration factor with 3 (three) concentration levels of 0.2 mM; 0.5 mM; and 0.8 mM. Incubation temperature variation factor (S) with three temperature levels of 250C, 30oC, and 350C. There are 9 different sample combinations including K1S1, K1S2, K1S3, K2S1, K2S2, K2S3, K3S1, K3S2, K3S3. Each treatment is repeated 3 (three) times. The results of the test will be analyzed using ANOVA and DMRT test. The results of the research conducted were the highest TGR values in samples with the addition of 0.8mM CuSO4 inducers of 10.645 mg / g. The highest TSP value was a sample with the addition of a 0.5mM CuSO4 inducer of 0.0343mg/g. The pH of the entire sample decreased (close to acidic) and the highest lignin removal value was 23.108% of the sample with the addition of 0.5mM CuSO4 inducer. CuSO4 inducers exert influence on delignification.
