Abstract
Pendahuluan: Diet tinggi kalori meningkatkan faktor risiko terjadinya obesitas. Pada obesitas, selain terjadi akumulasi lemak dalam jaringan termasuk paru yang mengakibatkan infiltrasi makrofag proinflamasi pada paru. Makrofag proinflamasi (M1) menginduksi glikolisis anaerob sedangkan makrofag M2 berperan dalam sekresi antiinflamasi pada adiposit. Metode: Penelitian ini menggunakan desain true eksperimental in vivo test, post test only control group pada 16 tikus Sprague Dawley (Rattus norvegicus) jantan usia 10-12 minggu yang terbagi dalam 2 kelompok, yaitu kelompok diet tinggi kalori, dan kelompok diet normal. Pemeriksaan akumulasi lemak paru melalui pewarnaan Oil Red O dan rasio M1/M2 jaringan paru melalui ekspresi CD11c dan CD206 dengan metode imunohistokimia. Uji statistik menggunakan uji deskriptif. Hasil: BMI kelompok diet tinggi kalori lebih tinggi dibandingkan kelompok diet normal (0,72±0,02 vs 0,62±0,03; p=0,000). Luas area akumulasi lemak paru kelompok diet tinggi kalori lebih tinggi dibandingkan kelompok diet normal (mean 24,12 vs 1860,77; p=0,001). Rasio makrofag M1/M2kelompok diet tinggi kalori lebih tinggi dibandingkan kelompok diet normal (mean 0,23 vs 0,832; p=0,000). Terdapat korelasi positif antara akumulasi lemak dengan rasio makrofag M1/M2 (r=0,936; p=0,000). Kesimpulan: Terdapat pengaruh diet tinggi kalori terhadap rasio makrofag M1/M2 melalui akumulasi lemak pada paru tikus. Semakin tinggi akumulasi lemak paru maka rasio makrofag M1/M2 akan semakin tinggi.
English Abstract
Introduction: High-calorie diet increases obesity risk that result in infiltration of proinflammatory macrophages caused by fat accumulation in tissues, including lung. M1 macrophages type as proinflammatory cells otherwise M2 macrophages as anti-inflammatory cells. This study analyses the effect of high-calorie diet on M1/M2 macrophages ratio through fat accumulation. Methods: It’s a true experimental in vivo test, post-test only control group in 16 male Sprague Dawley rats aged 10-12 weeks divided into 2 groups; high-calorie diet and normal diet group for 16 weeks. Fat accumulation examined by Oil Red O staining and M1/M2 ratio by expression of CD11c and CD206 by immunohistochemical method. Results: BMI of the high-calorie diet group was higher than the normal diet group (0.72±0.02 vs 0.62±0.03; p=0.000). Lung fat accumulation of the highcalorie diet group was higher than the normal diet group (32.73 ± 10.55 vs 0.37 ± 0.38; p=0.000). M1/M2 macrophages ratio of high-calorie diet group was higher than the normal diet group (0.831±0.20 vs 0.24±0.01; p=0.000). There is a significant positive correlation, very strong between fat accumulation and the ratio of M1/M2 macrophages (r=0.933; p=0.000). Conclusion: High-calorie diet increase BMI and fat accumulation in rats that shift M1/M2 ratio to proinflammatory (M1) type.
