Abstract
Kegiatan akuakultur memiliki potensi memajukan perekonomian yang ada di Indonesia. Akan tetapi kegiatan akuakultur sering kali mengalami permasalahan. Permasalahan yang sering muncul adalah munculnya penyakit akibat parasit, bakteri, virus, dan juga jamur. Salah satu bakteri yang sering menyerang kegaiatan akuakultur adalah bakteri A. hydrophila. Pengobatan komoditas yang terserang bakteri A. hydrophila umumnya menggunakan antibiotik, namun penggunaan antibiotik ini menjadikan bakteri menjadi lebih resisten. Oleh karena itu digunakanlah bahan alami yaitu daun pandan wangi (P. amaryllifolius Roxb) sebagai pengganti penggunaan antibiotik. Penelitian ini dilaksanakan di Laboratorium Budidaya Ikan Divisi Penyakit dan Kesehatan Ikan pada bulan Desember-Januari 2023. Tujuan penelitian ini adalah untuk mengetahui sensitivitas ekstrak daun pandan wangi (P. amaryllifolius Roxb) terhadap bakteri A. hydrophila secara in vitro. Metode penelitian ini adalah metode eksperimental. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan menggunakan 5 perlakuan, 3 ulangan dan 2 kontrol. Dosis ekstrak daun pandan wangi (P. amaryllifolius Roxb) yang digunakan yaitu A (30 ppm), B (60 ppm), C (90 ppm), D (120 ppm), dan E (150 ppm). Kontrol positif menggunakan antibiotik tetracycline 30 ppm dan kontrol negatif tanpa perlakuan. Hasil penelitian menunjukkan bahwa bakteri A. hydrophila sensitif terhadap setiap dosis ekstrak daun pandan wangi (P. amaryllifolius Roxb) yang diberikan. Hasil rerata diameter terbesar pada perlakuan dosis E (150 ppm) sebesar 11,03 ± 0,38 mm dan terkecil pada perlakuan dosis A (30 ppm) sebesar 8,36 ± 0,29 mm. Hubungan zona bening antar perlakuan ekstrak daun pandan wangi (P. amaryllifolius Roxb) terhadap bakteri A. hydrophila pola berbentuk linier dengan persamaan persamaan y =7,3267 + 0,0256x serta koefisien R2 = 0,843. Kesimpulan yang didapat bahwa daun panda wangi (P. amaryllifolius Roxb) berpengaruh nyata terhadap bakteri A. hydrophila. Bakteri A. hydrophila sesnsitif terhadap pemberian perlakuan ekstrak daun pandan wangi (P. amaryllifolius Roxb) dengan cara menghambat. Dosis perlakuan terbaik yaitu pada perlakuan E (150 ppm) dengan rerata yang didapat 11,03 mm.
English Abstract
Aquaculture activities have the potential to advance the existing economy in Indonesia. However, aquaculture activities often experience problems. The problem that often arises is the emergence of diseases caused by parasites, bacteria, viruses, and also fungi. One of the bacteria that often attacks aquaculture activities is A. hydrophila bacteria. Treatment of commodities attacked by A. hydrophila bacteria generally uses antibiotics, but the use of these antibiotics makes the bacteria more resistant. Therefore, natural ingredients are used, namely fragrant pandan leaves (P. amaryllyafolius Roxb) as a substitute for using antibiotics. This research was conducted in the fish farming laboratory, fish disease and health division in December-January 2023. The purpose of this study was to determine the sensitivity of Pandan Wangi leaf extract (P. amaryllifolius Roxb) to A. hydrophila bacteria in vitro. This research method is an experimental method. The experimental design used was a completely randomized design (CRD) using 5 treatments, 3 replications and 2 controls. The doses of Pandan Wangi leaf extract (P. amaryllifolius Roxb) used were A (30 ppm), B (60 ppm), C (90 ppm), D (120 ppm), and E (150 ppm). The positive control used 30 ppm tetracycline antibiotics and the negative control used no treatment. The results showed that A. hydrophila bacteria were sensitive to each dose of Pandan Wangi leaf extract (P. amaryllifolius Roxb) given. The largest mean diameter in the treatment dose E (150 ppm) was 11.03 ± 0.38 mm and the smallest in the treatment dose A (30 ppm) was 8.36 ± 0.29 mm. The clear zone relationship between pandan wangi leaf extract (P. amaryllifolius Roxb) treatment and A. hydrophila bacteria has a linear pattern with the equation y =7,3267 + 0,0256x and the coefficient R2 = 0,843. The conclusion obtained is that fragrant pandan leaves (P. amaryllifolius Roxb) have a significant effect on A. hydrophila bacteria. Bacteria A. hydrophila is sensitive to the treatment of pandan leaf extract (P. amaryllifolius Roxb) by inhibiting it. The best treatment dose was treatment E (150 ppm) with an average of 11.03 mm.
