Abstract

Latar Belakang : Insomnia didefinisikan sebagai kesulitan untuk memulai tidur, mempertahankan tidur, atau kondisi tidur yang tidak mampu merestorasi kondisi tubuh dan mental dengan adekuat. Namun terapi insomnia belum adekuat untuk memperbaiki kondisi insomnia. Terdapat beberapa pilihan terapi alternatif untuk terapi insomnia, misalnya salah satu yang terkenal adalah daun pegagan (Centella asiatica). Banyak jalur yang menyebabkan regulasi tidur, termasuk irama sirkardian dan reaksi seluler dimana terdapat fungsi apoptosis di dalamnya. Dibutuhkan penelitian untuk memperbaiki kondisi insomnia dengan pemberian Centella asiatica dengan pengamatan pada ikan zebra model insomnia. Tujuan Penelitian : Membuktikan pemberian ekstrak Pegagan (Centella asiatica) mempengaruhi jalur apoptosis melalui penghambatan ekspresi Apaf-1 dan Bax pada larva ikan zebra (Denio rerio) model insomnia. Metode dan Sampel Penelitian : Studi true laboratory experimental dengan pendekatan randomized control group post test only, dengan sampel 50 larva ikan zebra (Danio rerio) model insomnia yang diberikan perlakuan ekstrak pegagan (Centella asiatica) konsentrasi 2.5 μg/mL, 5 μg/mL dan 10 μg/mL yang dilakukan di Laboratorium Farmakologi, Laboratorium Patologi Anatomi, dan Laboratorium Biomedik Fakultas Kedokteran Universitas Brawijaya Malang selama 3 bulan, yakni pada bulan September hingga November 2022. Hasil Penelitian : Dari penelitian ini didapatkan bahwa terdapat pengaruh yang dari pemberian ekstrak pegagan terhadap ekspresi Apaf-1 (p=0.014), dengan ekstrak pegagan dengan konsentrasi 2.5 dan 5 μg/mL menurunkan ekspresi Apaf-1 pada kelompok larva ikan Zebra model insomnia (p=0.001, dan p=0.022). Terdapat pengaruh yang dari pemberian ekstrak pegagan terhadap ekspresi BAX (p=0.048) dengan ekstrak pegagan dengan konsentrasi 2.5 dan 5 μg/mL menurunkan ekspresi BAX pada kelompok larva ikan Zebra model insomnia (p=0.008 dan p=0.022). iv Kesimpulan : Pemberian ekstrak pegagan (Centella asiatica) pada konsentrasi 2.5 dan 5 μg/mL dapat menurunkan ekspresi Apaf-1 dan Bax pada larva ikan zebra model insomnia

English Abstract

Background : Insomnia is defined as difficulty falling asleep, maintaining sleep, or sleeping conditions that are unable to adequately restore the body and mental state. However, insomnia therapy has not been adequate to improve the condition of insomnia. There are several alternative therapy options for treating insomnia, for example one that is well known is gotu kola leaves (Centella asiatica). Many pathways lead to regulation of sleep, including circadian rhythms and cellular reactions in which apoptotic functions are involved. Research is needed to improve the condition of insomnia by administering Centella asiatica with observations on zebrafish models of insomnia. Objectives : Proving that administration of Centella asiatica extract influences the apoptotic pathway through inhibition of Apaf-1 and Bax expression in zebrafish larvae (Denio rerio) insomnia model. Material and Methods : A true laboratory experimental study with a randomized control group post test only approach, with a sample of 50 zebrafish larvae (Danio rerio) insomnia model that was given treatment with Centella asiatica extract at concentrations of 2.5 μg/mL, 5 μg/mL and 10 μg/mL which carried out at the Pharmacology Laboratory, Anatomical Pathology Laboratory, and the Biomedical Laboratory, Faculty of Medicine, Brawijaya University, Malang for 3 months, from September to November 2022. Results : From this study it was found that there was a significant effect of having Centella asiatica extract on the expression of Apaf-1 (p=0.014), with Centella asiatica extract at concentrations of 2.5 and 5 μg/mL reducing Apaf-1 expression in the group of zebrafish larvae with the insomnia model (p=0.001 , and p=0.022). There was a significant effect of having Centella asiatica extract on BAX expression (p=0.048) with Centella asiatica extract at concentrations of 2.5 and 5 μg/mL reducing BAX expression in the group of zebrafish larvae with the insomnia model (p=0.008 and p=0.022). vi Conclusion : Administration of Centella asiatica extract at a concentration of 2.5 and 5 μg/mL can reduce the expression of Apaf-1 and Bax in zebrafish larvae with insomnia models.
