Abstract
Penelitian ini terdiri atas dua tahap yaitu; Penelitian tahap 1 uji daya hambat temulawak enkapsulasi dalam menghambat pertumbuhan bakteri asam laktat (BAL), Salmonella sp dan Escherichia coli, dan penelitian tahap 2 uji biologis terhadap penampilan produksi (konsumsi pakan, pertambahan bobot badan, konversi pakan, persentase karkas, mortalitas dan Income Over Feed Cost/IOFC) dan karakteristik usus halus ayam pedaging (tinggi vili, jumlah vili, luas permukaan vili, kedalaman kripta, viskositas digesta dan pH usus). Rancangan percobaan yang di gunakan pada penelitian tahap 1 dan 2 adalah Rancangan Acak Lengkap (RAL). Penelitian tahap 1 terdiri atas 6 perlakuan dan 4 ulangan yaitu; T0- aquades, T1+ antibiotik 0,1%, T2 (temulawak enkapsulasi 25%), T3 (temulawak enkapsulasi 50%), T4 (temulawak enkapsulasi 75%) dan T5 (temulawak enkapsulasi 100%), dan penelitian tahap 2 terdiri atas 6 perlakuan dan 4 ulangan yaitu; ; P0- kontrol negatif, P1+ kontrol positif (antibiotik 0.1%), P2 (temulawak enkapsulasi 0.25%), P3 (temulawak enkapsulasi 0.50%), P4 (temulawak enkapsulasi 0.75%) dan P5 (temulawak enkapsulasi 1%). Data hasil penelitian dianalisa menggunakan Analysis of Variance (ANOVA) apabila ada perbedaan di antara perlakuan dilanjutkan dengan uji jarak Duncan’s Multiple Range Test (DMRT). Hasil penelitian tahap 1 aktivitas antibakteri temulawak enkapsulasi pada perlakuan T5 dapat menghambat bakteri Escherichia coli dan belum bisa menghambat bakteri asam laktat (BAL) dan Salmonella sp. Hasil penelitian menunjukkan perlakuan berpengaruh sangat nyata (P<0.01) terhadap bakteri Escherichia coli dan pengaruh tidak nyata (P>0.05) terhadap bakteri asam laktat dan Salmonella sp. Hasil penelitian tahap 2 menunjukkan temulawak enkapsulasi pada perlakuan P3 dapat meningkatkan bobot badan, persentase karkas, nilai IOFC, tinggi vili, jumlah vili, luas permukaan vili usus, dan menurunkan nilai konversi pakan, tingkat mortalitas dan pH usus. Penambahan temulawak enkapsulasi pada pakan ayam pedaging dapat memberikan pengaruh sangat nyata (P<0.01) terhadap IOFC, tinggi vili, jumlah vili, luas permukaan vili dan viskositas, dan berpengaruh nyata (P<0.05) terhadap bobot badan, konversi pakan, dan pH usus, serta memberikan pengaruh yang tidak nyata (P>0.05) terhadap konsumsi pakan, persentase karkas, mortalitas dan kedalaman kripta usus. Kesimpulan dari penelitian ini adalah temulawak enkapsulasi pada level 100% dapat menghambat pertumbuhan bakteri Escherichia coli, serta ditambahkan pada pakan ayam pedaging dengan level 0.5% direkomendasikan sebagai alternatif aditif pakan untuk menggantikan penggunaan antibiotik sintetik agar memperoleh penampilan produksi yang optimal.
English Abstract
This research consists of two experiments, namely; Phase 1 research testing the inhibition of encapsulated curcuma xanthorrhiza in inhibiting the growth of lactic acid bacteria (LAB), Salmonella sp and Escherichia coli, and Phase 2 research biological tests on production performance (feed consumption, body weight gain, feed conversion, carcass percentage, mortality and Income Over Feed Cost /IOFC) and characteristics of the small intestine of broilers (villi height, number of villi, villi surface area, kripta depth, digesta viscosity and intestinal pH). The experimental design used in phase 1 and 2 of the study was a completely randomized design (CRD). Phase 1 research consisted of 6 treatments and 4 replicates, namely; T0- distilled water, T1 + 0.1% antibiotic, T2 (25% encapsulated t curcuma xanthorrhiza), T3 (50% encapsulated curcuma xanthorrhiza), T4 (75% encapsulated curcuma xanthorrhiza) and T5 (100% encapsulated curcuma xanthorrhiza), and phase 2 research consisted of 6 treatments and 4 replicates, namely; P0- negative control, P1 + positive control (0. 1%), P2 (curcuma xanthorrhiza encapsulation 0.25%), P3 (curcuma xanthorrhiza encapsulation 0.50%), P4 (curcuma xanthorrhiza encapsulation 0.75%) and P5 (curcuma xanthorrhiza encapsulation 1%). Data from the study were analyzed using Analysis of Variance (ANOVA) if there were differences between treatments followed by Duncan's Multiple Range Test (DMRT). The results of the first stage of research on the antibacterial activity of encapsulated curcuma xanthorrhiza in the T5 treatment can inhibit Escherichia coli bacteria and cannot inhibit lactic acid bacteria (LAB) and Salmonella sp. The results showed that the treatment had a very significant effect (P <0.01) on Escherichia coli bacteria and no significant effect (P>0.05) on lactic acid bacteria and Salmonella sp. The results of stage 2 showed that the use of encapsulated curcuma xanthorrhiza in P3 treatment could increase body weight, carcass percentage, IOFC value, villi height, number of villi, intestinal villi surface area, and reduce feed conversion value, mortality rate and intestinal pH. The addition of encapsulated curcuma xanthorrhiza to broiler feed can have a very significant effect (P<0.01) on IOFC, villi height, villi number, villi surface area and viscosity, and has a significant effect (P<0.05) on body weight, feed conversion, and intestinal pH, and has no significant effect (P>0.05) on feed consumption, carcass percentage, mortality and intestinal krypta depth. The conclusion of this study is that encapsulated curcuma xanthorrhiza at 100% level can inhibit the growth of Escherichia coli bacteria, and added in broiler feed at a level of 0.5% and recommended as an alternative feed additive to replace the use of synthetic antibiotics to obtain optimal broiler production performance.
